Fertilidade e morfofisiologia epididimária de ratos wistar submetidos a ingestão de arsenato e arsenito de sódio

Resumo: The aim of this study was to compare the effect of exposure to different concentrations of arsenic in the pentavalent (arsenate, As+5) and trivalent forms (arsenite, As+3) on reproductive parameters. Were used 30 Wistar rats randomly divided into five experimental groups (n = 6 animals/group). The control animals received saline, while the treated animals were exposed orally to sodium arsenate and arsenite, testing for each chemical form with concentrations of 0.01mg/L and 10mg/L, daily for 56 days. Animals exposed to 10mg/L of arsenite had reduced daily sperm production, the number of spermatids in the testis and per gram of testis, the number of sperm in the epididymal regions and the percentage of sperm with intact membranes (P < 0.05). Already 0.01mg/L arsenite caused a reduction in the number of sperm in the caput/corpus regions of the epididymis and the percentage of sperm with intact membranes (P < 0.05). In addition, there was increase in the percentage of blood vessels in the initial segment of the epididymis to the animals exposed to arsenite (0.01 and 10mg/L) and arsenate (0.01mg/L). Sodium arsenate at the concentration to 10mg/L, only changed the percentage of sperm with intact membranes and percentage of lumen without sperm in the cauda of the epididymis (P < 0.05). Regarding the fertility parameters, there was obtained 81.82% of pregnant females to males control, 50% of males exposed with arsenate 0.01 mg/L and arsenite 10 mg/L and 63.64% of males exposed to 10mg/L of arsenate. 81.8% of the females conceived from males, 50% when the males were treated with 0.01 mg/L of arsenate and 10 mg/L of arsenite and 63.6% when 10 mg/L of arsenate were used. There was no difference between females groups for number of corpora lutea and implantations (P > 0.05), and the fertility potential (%) for the males exposed with arsenate 0.01mg/L (40.6 ± 12.8) and arsenite 10mg/L (40.6 ±11.7) was lower than the control males (71.0 ± 21.4). The pre-implantation loss was 59% for males exposed to arsenite 10mg/L and arsenate 0.01mg/L and 29% for control animals. In conclusion, the arsenite was more damaging than sodium arsenate some parameters for testicular and epididymal sperm. Utilizou-se 30 ratos divididos em cinco grupos (n= 6 animais/ grupo). Animais controle receberam NaCl a 0,9%, enquanto que os demais foram tratados diariamente por via oral com 0,01 e 10mg/L de arsênio, nas formas de arsenato e arsenito de sódio por 56 dias. Animais tratados com 10mg/L de arsenito tiveram redução na produção espermática diária, no número de espermátides no testículo e por grama de testículo, no número de espermatozoides nas regiões epididimárias e no percentual de espermatozoides com membranas íntegras (P < 0,05). Já 0,01mg/L de arsenito causou redução no número de espermatozoide nas regiões de cabeça/corpo do epidídimo e no percentual de espermatozoides íntegros (P < 0,05). Houve aumento no percentual de vasos sanguíneos no segmento inicial do epidídimo de animais tratados com arsenito (0,01 e 10mg/L) e arsenato (0,01mg/L). Arsenato de sódio, na concentração de 10mg/L, alterou o percentual de espermatozoides íntegros e o percentual de lúmen sem espermatozoides na cauda do epidídimo (P < 0,05). 81,8% de fêmeas conceberam de machos controle, 50% quando os machos foram tratados com arsenato 0,01 mg/L e arsenito 10 mg/L e 63,6% quando10mg/L de arsenato foi utilizado. Não houve diferença entre os grupos de fêmeas para número de corpos lúteos e de implantações (P > 0,05), sendo o potencial de fertilidade (%) de machos tratados com arsenato 0,01mg/L (40,6±12,8) e arsenito 10mg/L (40,6±11,7) menor que o dos machos controle (71,0±21,4). A perda embrionária pré-implantação foi de 59% para machos tratados com arsenato 0,01mg/L e arsenito 10mg/L e 29% para animais controle. Pode-se concluir que o arsenito foi mais danoso que o arsenato de sódio para parâmetros testiculares, epididimários e espermáticos.
