Estrutura e ultra-estrutura dos espermatozoides de Diaphorina citri (Hemiptera, Sternorryncha, Liviidae)

Resumo: The Hemiptera comprises a diverse group of insects with approximately 89,000 described species and distributed throughout the world. Currently, four suborders of Hemiptera are recognized: Heteroptera, Sternorryncha, Coleorryncha and Auchenorryncha. The suborder Sternorryncha form a monophyletic group with approximately 16,000 described species, distributed in four superfamilies: Aleyrodoidea, Aphidoidea, Coccoidea and Psyloidea. Some species of this suborder are among the major pests of agriculture worldwide. One example is Diaphorina citri Kuwayama (Liviidae), popularly known as the leafhopper, which today is one of the most important and devastating citrus pest worldwide. Considering knowledge about the reproductive biology as well as systematic may contribute to the management of pest, in this paper we describe the structure and ultrastructure of spermatozoa of D. citri. For this, were used light, transmission and scanning electronic microscopy. In this species we observed that the sperm measure about 525 μm in length, are free in the seminal vesicle and do not show polymorphism. As observed in other Psyloidea, the sperm when manipulated opens into two filaments, one in which its anterior end is attached to the nuclei and the other free. Through the flagellum there are lateral projections with about 2 μm in length, this characteristic is reported for another Psyloidea and Aleyrodoidea. Furthermore, at the end of the flagellum three appendages are observed with approximately 7 μm, which were observed so far only in D. citri. The head region is formed by nuclei with compact chromatin and a structure comprising by a matherial with median electron density and it extends extends about 25 μm in length in front of the nuclei. The flagellum consists of an axoneme (9 + 9 + 2), two mitochondrial derivatives and two accessory bodies. The presence of lateral projections is a characteristic shared by Psyloidea and Aleirodoidea, already the sperm opening in two filaments when manipulated occurs only in Psyloidea, and the presence of the three appendices at the end of the flagellum distinguishes D. citri from the other Psyloidea species studied.

A subordem Sternorryncha forma um grupo monofilético com aproximadamente 16 mil espécies descritas e distribuídas em quatro superfamílias: Aleyrodoidea, Aphidoidea, Coccoidea e Psylloidea. Algumas espécies dessa subordem estão entre as principais pragas da agricultura mundial. Um exemplo é a Diaphorina citri Kuwayama (Liviidae), popularmente conhecida como cigarrinha- do-limão, que hoje é uma das mais importantes e devastadoras pragas da citricultura mundial. Considerando que conhecer a biologia reprodutiva, bem como a sistemática, pode contribuir para o manejo de espécies-praga, neste trabalho descrevemos a estrutura e ultra-estrutura dos espermatozóides de D. citri. Para isso foram utilizadas microscopias de luz e eletrônicas de transmissão e varredura. Nesta espécie observamos que os espermatozoides medem cerca de 525 μm de comprimento, estão livres na vesícula seminal e não apresentam polimorfismo. Como observado em outros Psylloidea, o espermatozoide, quando manipulado, se abre em dois filamentos, onde um deles em sua extremidade anterior está preso ao núcleo e o outro livre. Ao longo de todo o flagelo existem projeções laterais com cerca de 2 μm de comprimento, característica descrita para outros Psylloidea e, também, Aleyrodoidea. Ainda, na extremidade final da cauda são observadas três apêndices de aproximadamente 7 μm, os quais foram observados até o momento apenas em D. citri. A região da cabeça é formada pelo núcleo com cromatina bastante compacta e por uma estrutura composta de material de densidade eletrônica mediana que se estende cerca de 25 μm de comprimento à frente do núcleo. O flagelo é constituído pelo axonema (9 + 9 + 2), dois derivados mitocondriais e dois corpos acessórios.
