Identificação e validação de seqüências expressas em tecido muscular de suínos

Resumo: The study of expressed sequences has great importance to know and understand the genetic architecture that control quantitative traits. In this way, in this study it was produced expressed sequence tags (ESTs) in muscular tissue of pigs from Large White and Piau breeds, these ESTs were compared with public data bases and differential expressed ESTs were identified between both libraries. With this objective it was generated two cDNA libraries from semimenbranosus muscle of adult (180 days) Large White and Piau pigs. In both libraries, 4,123 ESTs have been sequenced from 5 and 3 ends. After quality analysis, 1030 sequences were considered valid for the further analysis, according to established parameters. From this total, 675 ESTs were originated from Large White library and 355 ESTs were originated from Piau library. The analyzed ESTs presented average length of 494.76 pb. Cluster analysis by CAP3 software indicates that a total of 130 sequences (11.80%) was assembled in contigs and 900 ESTs (88.20%) represented unique sequences in this collection. The sequences were compared with nucleotide and protein public data bases, resulting in 333 homologues sequences (31.54%) and 697 (68.46%) sequences that did not present homology. A total of 255 ESTs (24.30%) was submitted to genomic annotation according to Gene Ontology Consortium. In this way, the ESTs were classified according to their molecular function, process and component. Differential expressed sequences were identified between both cDNA libraries. Most of the expressed sequences represent genes not previously described in muscle tissue of pigs.

Dessa forma, nesse estudo geraram-se etiquetas de seqüências expressas (ESTs) no tecido muscular de suínos das raças Large White e Piau, estas foram comparadas com seqüências depositadas em bancos de dados públicos e identificaram-se ESTs diferencialmente expressas entre ambas as bibliotecas. No intuito de identificar genes expressos na musculatura, foram construídas duas bibliotecas de cDNA a partir do músculo semimembranosus de animais adultos (180 dias) das raças Large White e Piau. Um total de 4.123 ESTs foi seqüenciado a partir das extremidades 5 e 3 , em ambas as bibliotecas. Após a análise de qualidade, 1030 seqüências foram validadas segundo os parâmetros estabelecidos para análises posteriores. Desse total, 675 ESTs pertenciam à biblioteca Large White e 355 ESTs pertenciam à biblioteca Piau. As ESTs validadas apresentaram tamanho médio de 494,76 pb. As seqüências foram então submetidas à análise de agrupamento pelo programa CAP3 resultando em 130 seqüências (11,80%) que puderam ser agrupadas em contigs e 900 ESTs (88,20%) se apresentaram como seqüências únicas. As seqüências foram comparadas com bancos de dados de nucleotídeos e proteínas, resultando em 333 seqüências (31,54%) homólogas e 697 seqüências (68,46%) sem homologia. Um total de 255 ESTs (24,30%) foi submetido à anotação gênica segundo as normas do consórcio Gene Ontology. Dessa forma, as ESTs foram classificadas quanto à função molecular, ao componente celular e ao processo biológico no qual estavam envolvidas. Foram identificadas seqüências diferencialmente expressas entre as bibliotecas de cDNA analisadas.
