Genetic variability through isozimic polymorphisms in hexaploid oats

Resumo: Analysis of isozymes have many applications in plant breeding specially in the study of genetic variability. Isozymes are direct product of genic expression and may help to understand the evolutionary relation between species, and allow to identify the potential genotypes for breeding programs. This work aimed to identify genetic variability through esterase polymorphism and to estimate genetic similarity among three hexaploid oat groups. Nine cultivated oat genotypes, three introductions of A. sterilis L., and 16 introductions of A. fatua L. were used. Wide polymorphisms of isozyme were observed within and between studied groups. The electrophoretic analysis showed 13 different bands with anodical migration between the genotypes. Although several bands were common for all genotypes, some of them were typical for a particular hexaploid group. These results indicate high genetic similarity between cultivated oat and A. fatua group. The A. sterilis group was more divergent, due to a more differentiated electrophoretic pattern.

The A. sterilis group was more divergent, due to a more differentiated electrophoretic pattern.A análise isoenzimática por eletroforese possui várias aplicações no melhoramento vegetal, principalmente na identificação de variabilidade genética – pois as isoenzimas são produtos diretos da expressão gênica –, e ajuda a entender a relação evolutiva entre as espécies, possibilitando a identificação de genótipos potenciais a serem utilizados nos programas de melhoramento. O presente trabalho teve por objetivo identificar a variabilidade genética através dos polimorfismos do sistema isoenzimático esterase e estimar a similaridade genética entre três grupos hexaplóides de aveia. Para a análise foram utilizados nove genótipos cultivados de aveia, três introduções silvestres de Avena sterilis L., e dezesseis introduções silvestres de A. fatua L. Ampla variabilidade genética de polimorfismos isoenzimáticos foi observada dentro e entre os grupos avaliados. A análise eletroforética revelou treze bandas diferenciais com migração anódica entre os genótipos testados. Apesar de existirem bandas isoesterásicas comuns entre eles, algumas delas parecem ser características de determinado grupo hexaplóide.
