Immunotherapies with immune checkpoint inhibitors (ICIs) are becoming a promising tool to replace conventional cancer therapies. These ICI-based immunotherapies have shown remarkable therapeutic success in various cancers, but their efficacy in GBM has yet to be shown. Increasing evidence indicates that aberrant WNT signalling, as what happens in GBM, promotes immunoevasion and resistance to ICIs. We recently established WNT6 as an oncogenic biomarker in GBM, which activates several key signalling pathways, including the WNT pathway. Interestingly, WNT6 expression is correlated with key immune system-related genes (e.g., CTLA-4 and PDL1) in GBM. This project aimed to decipher the impact of WNT6 in the immune response in GBM and evaluate the potential of combining targeted therapies (targeting WNT6 or WNT ligands) and ICIs. In this context, using WNT6 silencing and over-expression human GBM cell lines, we found that WNT6 expression is associated with IL1B expression in U373 and SNB19 cell lines, and with TNFA, IL8, TGFB1 and CCL2 expression in U373 cell line. Furthermore, WNT6 expression is inversely associated with IL6 and CCL2 in the SNB19 cell line. At protein levels, WNT6 affected the expression of PD-L1 and PD-L2, but the effect was cell line-dependent. We further demonstrated that WNT6 silencing significantly increases the sensitivity of SNB19 GBM cells to ICIs therapy, namely using anti-PD-1 and anti-CTLA-4 agents. In the same in vitro co-cultures, T-cell apoptosis and subpopulations (CD4+ , CD8+ and Tregs) were evaluated. Regarding this part, no differences were found in all human GBM cell lines tested, both at naïve and treatment conditions. Furthermore, the combination of ICIs with LGK974, a WNT ligands inhibitor, did not impact tumour cell apoptosis, showing low therapeutic potential for the targeting of GBM. Finally, we showed WNT6 has the same functional characteristics in a murine GBM cell line as those observed in human models. Overall, our findings indicate that WNT6 may affect the expression of several key cytokines in GBM, and impact ICIs efficacy in vitro. Nevertheless, to further clarify the present results, it will be vital to evaluate the secretion of these cytokines and chemokines, and to perform in vivo experiments with both immunocompetent and humanized GBM mice models to clarify the potential of combining WNT6 targeting with ICIs and WNT6 role in immune cells.O glioblastoma (GBM) é o tumor mais maligno do sistema nervoso central em adultos, sendo inevitavelmente fatal. As imunoterapias com inibidores de “checkpoints” imunes (ICIs) estão a tornar-se uma ferramenta promissora para complementar ou substituir as terapias convencionais utilizadas em cancro. O WNT6 foi descrito como um novo biomarcador oncogénico em GBM, ativando várias vias de sinalização oncogénicas, entre elas a via WNT. Curiosamente, em GBM, a expressão do WNT6 está positivamente correlacionada com genes relacionados com respostas imunes (por exemplo, CTLA-4 e PDL1). Neste contexto, este projeto teve como principais objetivos decifrar o impacto do WNT6 na resposta imune em GBM, e avaliar o potencial da combinação de terapias dirigidas (atuando ao nível do WNT6 ou da via WNT) e ICIs, utilizando linhas celulares humanas de GBM, manipuladas para sobreexpressar ou silenciar o WNT6. Primeiramente, verificamos que a expressão do WNT6 influencia a expressão de IL1B nos dois modelos celulares de silenciamento do WNT6 (SNB19 e U373), e também a expressão de TNF, IL8, TGB1 e CCL2 na linha U373. Na linha celular SNB19, a expressão do WNT6 está inversamente relacionada com a expressão de IL6 e CCL2. Ao nível da proteína, o WNT6 afeta os níveis de PDL1 e PLD2 na membrana celular, apesar deste efeito ser dependente da linha celular. Seguidamente, demostramos que o silenciamento do WNT6 aumenta a sensibilidade das células tumorais à terapia com ambos ICIs, anti-CTLA-4 e anti-PD-1, na linha celular SNB19. Nas mesmas co culturas, a apoptose e as subpopulações de células T foram avaliadas, mas não foram encontradas diferenças em qualquer das condições testadas. A combinação de ICIs e LGK974, um inibidor da secreção de ligandos WNT, não teve um impacto significativo na morte das células tumorais, demostrado pouco potencial terapêutico no regime testado. Finalmente, manipulamos os níveis de WNT6 numa linha celular de GBM de ratinho (GL261) e descobrimos que o WNT6 potencia as propriedades oncogénicas destas células de GBM. Globalmente, os nossos resultados demostraram que o WNT6 afeta a expressão de várias citoquinas e quimocinas em GBM, e a sensibilidade a ICIs in vitro. Futuramente, será de enorme relevância avaliar a secreção destas citocinas e quimocinas, e realizar experiências com modelos in vivo de GBM, de forma a clarificar o papel do WNT6 no sistema imune e a avaliar o potencial terapêutico da combinação de ICIs e terapias que tem como alvo o WNT6 no tratamento desta doença fatal.The work presented in this thesis was performed in the Life and Health Sciences Research Institute (ICVS), Minho University.
