Resumo: | Neuroblastoma (NB) is a malignancy neoplasm in the nervous system that almost exclusively occurs during infancy and early childhood. NB is responsible for 8% to 10% of cases of cancer in children. More than 50% of the diagnosed cases have generalized metastatic disease at the time of diagnosis. Trace elements elements are extremely important in the functioning of cellular processes, incluiding metabolic reactions and gene expressions. The Micro X-ray fluorescence (microXRF) technique has been widely used in biological sample analysis because it is a non-destructive technique, does not produce thermal damage and allows the analysis of several elements simultaneously. The objectives of this study were to obtain the elemental composition and elemental concentration presents in human neuroblastoma (SH-SY5Y and SK-NBE (2)). In addition, determine the limits detection of the microXRF system and analyze the spatial distributions (bidimensional maps) of the elements presents in samples comparing the NB tumor lines. Cellular spheroids were grown from 96-well plate culture cell using DMEM culture medium supplemented with 10% SFB for 5 days incubation. The microXRF spectra were obtained through the M4 Tornado spectrometer (Bruker Nano GmbH), being possible to detect the following elements: P, S, K, Cl, Ca, Fe, Cu and Zn. From a certified sample it was possible to calculate the limit detection of the system. The limit detection of the system ranged from 0.8 µg.g⁻¹ (for copper and zinc) to 7.2 µg.g⁻¹ (for phosphorus). The bidimensional maps of the chemical elements obtained showed non-uniform distributions for all elements and spheroids analyzed. In addition, the results obtained through quantification showed that the values obtained for the elementary concentrations showed great dispersion due to the heterogeneity of the cellular spheroids. The results showed through the nonparametric Mann-Whitney test with a significance level of 5% that the cell lines present significant differences. |

O NB acomete cerca de 8 % a 10 % dos casos de neoplasias em crianças, onde mais de 50 % dos casos apresentam doença metastática generalizada no momento do diagnóstico. Os elementos químicos são extremamente importantes no funcionamento de processos celulares, como reações metabólicas e expressões gênicas. A técnica de microfluorescência fluorescência de raios X (microXRF) tem sido amplamente empregada em análises de amostras biológicas por ser uma técnica não destrutiva, produzir dano térmico quase desprezível e permitir a análise de diversos elementos simultaneamente. O objetivo deste estudo foram obter as concentrações elementares presentes em esferoides de linhagens celulares de neuroblastoma humano (SH-SY5Y e SK-N-BE (2)). Além disso, determinar os limites de detecção do sistema de microXRF e analisar as distribuições espaciais (mapas bidimensionais) dos elementos presentes nas amostras comparando as linhagens tumorais de NB. Os esferoides celulares foram formados a partir de cultura celular em placa de 96 poços utilizando meio de cultura DMEM suplementado com 10% de SFB durante 5 dias de incubação. A análise por microXRF foi realizada através do espectrômetro M4 Tornado (Bruker Nano GmbH), sendo possível a detecção dos seguintes elementos: P, S, Cl, K, Ca, Fe, Cu e Zn. A partir de uma amostra certificada foi possível calcular o limite de detecção do sistema, estando este entre 0,8 µg.g⁻¹ (para cobre e zinco) até 7,2 µg.g⁻¹ (para fósforo). Os mapas bidimensionais dos elementos químicos mostraram distribuições não uniformes para todos os elementos e esferoides analisados. Além disso, os resultados obtidos através da quantificação mostraram que os valores obtidos para as concentrações elementares variaram desde valores mínimos de 1,5 µg.g⁻¹ até 3,0 µg.g⁻¹ para o elemento Zn e valores mínimos de 1064,0 µg.g⁻¹ até 2413,0 µg.g⁻¹ para o elemento P, respectivamente para as linhagens SK-N-BE (2) e SHSY5Y. Os resultados apresentaram grande dispersão devido a heterogeneidade dos esferoides celulares.
