Resumo: | The aim of this study was to investigate the effects of low-level red laser on tissue repair in rats submitted to second degree burn, evaluating if the timing of laser treatment influence on the healing process and investigate the expression of the genes involved with DNA repair by base excision and nucleotide excision pathways. The animals had their backs shaved and divided as follows: control group (n = 12) - rats burned and not irradiated, early laser group (n = 12) - rats burned and irradiated from day 1 after injury for 5 consecutive days, and late laser group (n = 14) - rats burned and irradiated from day 4 after injury for 5 consecutive days. Laser irradiation was according to a clinical protocol (20 J/cm2, 100 mW, continuous wave emission mode, 660 nm) as recommended by the laser device manufacturer. Half of the animals were sacrificed 10 days after burn and the other animals were sacrificed 21 days after burn. Late laser group accelerated wound contraction 10 and 21 days after burn. Late laser group accelerated reepithelialization 18 days after burn. Late laser group increased the granulation tissue 10 and 21 days after burn. Both irradiated groups increased type III collagen expression and TGF-β 21 days after burn. Both irradiated groups increased macrophages and myofibroblasts number 10 days after burn, and decreased 21 after days. Low-level red laser exposure reduced the expression of APE1 messenger (mRNA), increased the expression of OGG1, reduced the expression of XPC and increased the expression of XPA in both irradiated groups laser and later laser groups. Low-level red laser exposure contributes to the process of tissue repair of second degree burns, suggesting that the intervention during proliferative phase is crucial in the final outcome of the repair process. Moreover, the therapeutic laser was capable of altering the expression of genes involved in DNA repair. |

Moreover, the therapeutic laser was capable of altering the expression of genes involved in DNA repair.O objetivo do estudo foi investigar os efeitos do laser terapêutico vermelho de baixa potência no processo de reparo tecidual em ratos submetidos à queimadura de segundo grau, investigando ainda se o momento da intervenção pode influenciar influência no processo de cicatrização e a expressão de genes envolvidos no reparo do DNA pelos mecanismos de excisão de bases e de nucleotídeos. Os animais foram divididos da seguinte forma: grupo controle (n = 12) ratos submetidos à queimadura e não irradiados, grupo laser inicial (n = 12) ratos queimados e irradiados 1 dia após a queimadura e irradiados por 5 dias consecutivos e grupo laser tardio (n = 14) ratos queimados e irradiados 4 dias após a queimadura e irradiados por 5 dias consecutivos. A irradiação com laser foi baseada em protocolo clínico (20 J/cm2, 100 mW, modo contínuo, 660 nm) sugerido pelo fabricante do equipamento. Parte dos animais foi sacrificado 10 dias após a queimadura e a outra parte foi sacrificada 21 dias após a queimadura. O laser terapêutico acelerou o fechamento da lesão no grupo laser tardio 10 e 21 dias após a queimadura e acelerou a reepitelização 18 dias após a queimadura. Observou-se um aumento do tecido de granulação no grupo laser tardio 10 e 21 dias após a queimadura. Um aumento da expressão do colágeno tipo III e TGF-β foi observado 21 dias após a queimadura em ambos os grupo irradiados. Ambos os grupos irradiados tiveram aumento no número de macrófagos, miofibroblastos 10 dias após a queimadura e diminuição 21 dias após a queimadura. Quanto ao reparo do DNA, 10 dias após a queimadura, foi observado que o laser terapêutico aumenta a expressão de OGG1 e XPA reduz a expressão de APE1 e XPC em ambos os grupos irradiados. A exposição ao laser terapêutico vermelho de baixa potência contribuiu para o processo de reparo tecidual na queimadura de segundo grau, sugerindo que a intervenção durante a fase proliferativa é crucial no resultado final do processo de reparo.
