Resumo: | The relationship between production of reactive oxygen species and the development of atherosclerosis has been studied for decades. It is known that all inflammatory processes involved in plaque progression leads to increased production of free radicals. Furthermore, aging is a factor that aggravates atherosclerotic disease and also reactive oxygen species production, probably by the loss of antioxidant capacity that makes regular balancing of reactive oxygen species. The Apolipoprotein E deficient mouse is a model widely used in the study of atherosclerosis, in which there is high hypercholesterolemia that results in increased production of reactive oxygen species in the bone marrow, blood and aorta cells. Aging in turn also leads to increased production of reactive oxygen species, however when combined with hypercholesterolemia, the result of this phenomenon is the occurrence of increased production of reactive oxygen species and thus high levels of oxidative DNA fragmentation and apoptosis. In this regard, most notably is the analysis of bone marrow cells, a source of blood and stem cells for all organism that commonly presents a low oxygen tension ambient, in which we showed high generation of superoxide radicals and hydrogen peroxide. Thus, bone marrow is affected by oxidative imbalance caused by aging and atherosclerosis. Still, corroborating these data, in the analysis of DNA fragmentation by comet assay, bone marrow cells showed high levels of fragmentation in atherosclerotic and aged animals. Still, blood and aorta cells from atherosclerotic and aged animals also showed high superoxide anion production. However, there are low hydrogen peroxide production in endothelial cells of all groups, suggesting a possible modulation of tissue-specific effects of antioxidant enzymes. Furthermore, we note that there is not increased production of hydrogen peroxide in blood cells from aged C57 animals, unlike bone marrow mononuclear cells, in which we observed an increase. Finally, it can be noted that, in general, atherosclerosis increases the production of reactive oxygen species as well as aging, but when these two parameters are combined oxidative stress is further increased. The analysis of scientific literature suggests a possible activation of NADPH oxidase enzyme, responsible for the increased production of reactive oxygen species, and downregulation of antioxidant enzymes, which affect the balance of oxidative stress in tissues. |

Sabe-se que todo processo inflamatório envolvido na progressão da placa acarreta em aumento da produção de radicais livres. Além disso, o envelhecimento é um fator que agrava a doença aterosclerótica e também a produção de espécies reativas de oxigênio, provavelmente pela perda da capacidade antioxidante que faz o balanceamento regular das espécies reativas de oxigênio. O camundongo Apolipoproteína E deficiente é um modelo amplamente utilizado no estudo da aterosclerose, no qual há elevada hipercolesterolemia o que acarreta em aumento da produção de espécies reativas de oxigênio na medula óssea, sangue e aorta. O envelhecimento por sua vez também leva ao aumento da produção das espécies reativas de oxigênio, entretanto quando cumulado com a hipercolesterolemia, o resultado desse fenômeno é a ocorrência de maior produção de espécies reativas de oxigênio e, consequentemente, níveis elevados de fragmentação oxidativa do DNA e apoptose celular. Neste sentido, com maior destaque está a análise das células de medula óssea que apresentaram neste estudo alta produção de ânions superóxido e de radicais de peróxido de hidrogênio, ou seja, a fonte de produção de células sanguíneas e células tronco para todo o organismo, que comumente apresenta ambiente com baixa tensão de oxigênio, é afetada pelo desequilíbrio oxidativo causado pelo envelhecimento e pela aterosclerose. Ainda, corroborando com estes dados, a análise da fragmentação do DNA, pelo ensaio do cometa, das células de medula revelaram altos índices de fragmentação nos animais ateroscleróticos e idosos. Ainda, as células de sangue e aorta dos animais ateroscleróticos e idosos também apresentaram alta produção de ânion superóxido. Entretanto, encontramos baixa produção de peróxido de hidrogênio nas células endoteliais de todos os grupos, sugerindo uma possível modulação tecido-específica dos efeitos de enzimas antioxidantes. Além disso, observamos que não há aumento da produção de peróxido de hidrogênio nas células sanguíneas de animais C57 idosos, ao contrário das células mononucleares de medula óssea, no qual observamos aumento. Por fim, pode-se destacar que, de modo geral, a aterosclerose aumenta a produção de espécies reativas de oxigênio assim como o envelhecimento, porém quando estes dois parâmetros são cumulados a tensão oxidativa é ainda maior. A análise da literatura científica sugere uma possível ativação da enzima NADPH oxidase, responsável pelo aumento da produção de espécies reativas de oxigênio, e downregulation de enzimas antioxidantes, que prejudica o balanceamento do estresse oxidativo nos tecidos analisados.
