A influência do estresse ambiental no perfil lipídico e glicêmico de ratos Wistar tratados com prednisona

Resumo: The environment in which the individual is is directly related to quality of life. An inappropriate environment can lead to stress, in this situation there is the release of cortisol causing several effects in the body. Prednisone is a synthetic glucocorticoid, widely used in clinical practice once it has anti-inflammatory and immunosuppressive action, but depending on the time of treatment and dose used, it can cause changes in lipid metabolism and hyperglycemia. Thus, the objective of this study was to evaluate the influence of the stressful environment (metabolic cage) on the lipid and glycemic profile of Wistar rats treated with prednisone. For this, male Wistar rats were divided into groups: Cage with Prednisone (CP, n=7) and Metabolic Cage with Prednisone (MCP, n=7), treated daily with prednisone dissolved in the SyrSpend® vehicle, at a dose of 5mg/Kg /day for 28 days. The leukogram was evaluated before the beginning of treatment (day 0) and last day (28). On the 23rd day, the Oral Glucose Tolerance Test (OGTT) was performed, as well as the Area Under the Curve (AUC) and the Glucose Rate per Minute (GRM). On the 26th day, the Insulin Tolerance Test (ITT) was performed, as well as the area under the TTI curve and the Glycemic Uptake Index (GUI). At the end of the treatment, the rats were anesthetized and killed for analysis of serum biochemical parameters. Data were analyzed statistically, with significance at p<0.05. Comparing to the beginning of treatment, on the 28th day the segmented neutrophils of the GP group was increased and the eosinophils of the CP group decreased. In OGTT, the glycemic value of the GP group at 30' was higher than the CP group. As in the AUC of the OGTT and its TGM, it was verified that the glycemia of the GP rats was higher in the 30-60' time interval. Regarding serum lipids, there were no differences between groups; however, the total cholesterol and triglycerides values of both groups were found to be elevated to the reference values. It is concluded that the treatment with prednisone associated with stress interfered in the metabolism of glucose and lipids, as well as in the leukocyte profile.

Um ambiente não adequado pode levar ao estresse, nessa situação há a liberação do cortisol provocando diversos efeitos no organismo. A prednisona é um glicocorticoide sintético, muito utilizado na prática clínica por possui ação anti-inflamatória e imusossupressora, mas dependendo do tempo do tratamento e dose utilizados, pode causar alterações no metabolismo lipídico e hiperglicemia. Desta forma, o objetivo desse estudo foi avaliar a influência do ambiente estressor (gaiola metabólica) no perfil lipídico e glicêmico de ratos Wistar tratados com prednisona. Para isso, foram utilizados ratos Wistar machos divididos em grupos: Caixa com Prednisona (CP, n=7) e Gaiola com Prednisona (GP, n=7), tratados diariamente com prednisona dissolvida no veículo SyrSpend®, na dose de 5mg/Kg/dia por 28 dias. Foi avaliado o Leucograma antes do ínicio do tratamento (dia 0) e último dia (28). No 23 dia foi realizado o Teste Oral de Tolerância à Glicose (TOTG), bem como a Área sob a curva (ASC) e a Taxa de glicose por minuto (TGM). No 26 dia realizou-se o Teste De Tolerância à Insulina (TTI), assim como à área sob a curva do TTI e o Índice de Captação Glicêmica (ICG). Ao final do tratamento os ratos foram anestesiados e mortos para análise dos parâmetros bioquímicos séricos. Os dados foram analisados estatisticamente, com significância em p<0,05. Comparados ao início do tratamento, no 28º dia os neutrófilos segmentados do grupo GP aumentaram e os eosinófilos do grupo CP reduziram. No TOTG, o valor glicêmico do grupo GP aos 30’ foi maior do que o grupo CP. Assim como na ASC do TOTG e seu TGM, foi verificado que a glicemia dos ratos GP foi maior no intervalo 30-60’. Em relação aos lipídios séricos, não houve diferenças entre os grupos; entretanto, os valores de colesterol total e triglicerídeos de ambos os grupos se encontraram elevados aos valores de referência.
