Defesa : Curitiba, 08/03/2019Inclui referências: p. 99-112Resumo: A família Lauraceae e o gênero Ocotea são conhecidos por suas propriedades biológicas e farmacológicas. O estudo de galhas de Ocotea puberula (RICH.) Nees, contribui com a pesquisa de metabólitos secundários para atividades aplicadas às ciências farmacêuticas, agronômicas e química. A espécie é uma planta heliófita. O objetivo deste trabalho foi realizar o estudo do perfil alcaloídico e avaliar as propriedades antioxidantes e atividades biológicas, nas galhas dos frutos da espécie. A partir da maceração em álcool deu-se origem a fração extrato bruto que foi submetido a pricipitação à frio com propósito de separar a goma, gerando posteriormente o extrato sem goma e extrato goma. Os extratos foram submetidos à análise do perfil metabólico por CCD e se obteve resultados positivos para esteroides e triterpenos, compostos fenólicos e alcaloides. Para dosagem da dicentrina foram realizadas extrações alcaloídicas no extrato bruto, extrato sem goma, extrato goma, galhas, frutos e folhas normais. A fração com a maior quantificação de dicentrina foi à goma seguida de extrato bruto e extrato sem goma. A partir da fração goma obteve-se um precipitado que foi isolado e identificado como dicentrina. O extrato bruto, extrato sem goma, extrato goma foram submetidos aos testes antioxidantes por redução do complexo fosfomolibdenio e redução do radical DPPHo. No teste de DPPHo a fração goma apresentou IC50 (10,39 ?g/mL) e o extrato bruto IC50 (11,39 ?g/mL) estatisticamente iguais aos padrões BHT e rutina. O extrato bruto, extrato sem goma e goma apresentaram alto teor de compostos fenólicos. No ensaio de toxicidade in vitro frente à Artemia salina o isolado dicentrina foi considerado moderadamente tóxico. Na alelopatia, houve inibição do crescimento do hipocótilo no extrato bruto que inibiu nas concentrações 500, 750, e 1000 ?g/mL. No extrato sem goma, extrato goma e dicentrina inibiram o crescimento do hipocótilo nas concentrações de 250 até 1000 ?g/mL. Na radícula houve inibição na fração goma nas concentrações 500 e 750 ?g/mL, no extrato bruto 500, 750 e 1000 ?g/mL, dicentrina 750 e 1000 ?g/mL e extrato sem goma 250 até 1000 ?g/mL. Obteve-se atividade hemolítica na faixa de 30% a 50% na concentração de 100 ?g/mL da fração goma. Considerando a presença do constituinte dicentrina, as propriedades tóxicas e alelopática demonstradas pelos extratos, estudos de aplicações diversas podem sem aprofundados e direcionando novos estudos científicos. The Gallblot Study of Ocotea puberula (RICH.) Nees, contributes to the research of secondary metabolites for activities applied to the pharmaceutical, agronomic and chemical sciences. The species is a heliophyta plant. The objective of this work was to study the alkaloid profile and to evaluate the antioxidant properties and biological activities in the gall of fruits of the species. From the maceration in alcohol gave rise to the fraction crude extract that was subjected to cold pricipitation with the purpose of separating the gum, later generating the extract without gum and extract gum. The extracts were submitted to the analysis of the metabolic profile by CCD and positive results were obtained for steroids and triterpenes, phenolic compounds and alkaloids. For the dosage of dicentrin, alkaloid extractions were performed in the crude extract, extract without gum, extract gum, galls, fruits and normal leaves. The fraction with the highest quantification of dicentrin was to the gum followed by crude extract and extract without gum. From the gum fraction a precipitate was obtained which was isolated and identified as dicentrin. The crude extract, gumless extract, gum extract were submitted to the antioxidant tests by reduction of the phosphomolibdenium complex and reduction of the radical DPPH. In the DPPH test, the gum fraction had IC 50 (10.39 ?g / mL) and crude extract IC 50 (11.39 ?g / mL) statistically equal to the BHT and routine standards. The crude extract, extract without gum and gum presented high content of phenolic compounds. In the in vitro toxicity test against Artemia salina the isolated dicentrin was considered moderately toxic. In allelopathy, hypocotyl growth inhibition was inhibited in the crude extract which inhibited the concentrations at 500, 750, and 1000 ?g / mL. In the extract without gum, extract gum and dicentrin inhibited the growth of hypocotyl in the concentrations of 250 to 1000 ?g / mL. In the radicle there was inhibition in the 500 and 750 ?g / mL concentrations in the crude extract 500, 750 and 1000 ?g / mL, dicentrin 750 and 1000 ?g / mL and the extract without gum 250 to 1000 ?g / mL. Hemolytic activity was obtained in the range of 30% to 50% in the concentration of 100 ?g / mL of the gum fraction. Considering the presence of the constituent dicentrin, the toxic and allelopathic properties demonstrated by the extracts, studies of several applications can without in depth and directing new scientific studies.
