Resumo: | Objective: The objective of this study was to analyze the metabolic and behavioral effects of fetal growth restriction (IUGR) caused by the deficiency of protein during intrauterine life before and after exposure to high-fat diet. Method: After protein restriction (5g protein / 100 g diet, HP diet) in the maternal diet offspring, it was compared with offspring of mothers receiving normal protein diet (20 g protein / 100 g diet, NP diet). After birth, all offspring were suckled by progenitors fed NP diet and, there was adoption of all litters by progenitors fed NP diet. In postnatal day (PND) 28 a total of 51 offspring were distributed into two groups according to the maternal diet, namely: control group (CT, HP maternal diet) and restriction group (RT, NP maternal diet). The offspring were accompanied for 2 months receiving NP diet and, after this period all of them were fed with a high-fat diet with high concentration of saturated fat (31g saturated fat, 100g diet, HL) for about 5 weeks. The body weights of the offspring were accompanied weekly from birth to the 1st month of life and, monthly from 1st month until euthanasia. The naso-anal length was measured in PND30 and PND60. Blood samples were collected in PND30, PND60 and PND90 to assess blood concentrations of glucose, total cholesterol and triglycerides in whole blood and plasma concentrations of insulin and leptin. The animals were subjected to behavioral tests: Novelty suppressed-feeding test and open field. After behavioral assessments, the animals were euthanized by decapitation and had blood, fat, liver, kidney and muscle collected. Data is presented as mean ± standard deviation. Results and conclusions: IUGR decreased the birth weight of offspring and this effect can be observed at other times throughout the study; in males, this restriction reduced the naso-anal growth and altered muscle mass and the proportion of fat after high fat diet consumption. After spending the period stipulated feeding only at HL diet, restricted animals showed elevated plasma insulin. Males and females RT group showed increased locomotor activity, restricted females also had fewer episodes of self-cleaning during behavioral 95 testing and showed a higher consumption of HL diet over time in relation to the females CT group. With these data, we can conclude that the deficiency in maternal nutrition delayed intrauterine growth, and this effect can interfere in the post-natal development, affecting morphometric parameters before and after consumption of a HL diet. After passing through metabolic challenge exposure (HL diet), restricted animals showed elevated plasma insulin, demonstrating that IUGR may influence metabolic signaling involved in energy homeostasis promoted by a nutritional imbalance. |

Método: Camundongos fêmeas da linhagem Swiss foram submetidos a uma dieta controle (20g de proteína/100g de dieta normoproteica; NP) e experimental (5g de proteína/100g de dieta hipoproteica; HP) durante 14 dias antes do acasalamento e toda gestação. Após o nascimento, os filhotes HP foram adotados e amamentados por progenitoras do grupo controle, que receberam dieta NP. Para minimizar possível viés, filhotes controles foram randomizados e adotados por mães que receberam dieta NP. No dia pós-natal (DPN) 28 um total de 51 filhotes foram distribuídos em dois grupos experimentais de acordo com a dieta materna, a saber: grupo controle (CT, dieta materna HP) e grupo restrição (RT, dieta materna NP). A prole foi acompanhada por 2 meses recebendo dieta NP, após esse período foi ofertada a todos os animais uma dieta hiperlipídica com alta concentração de gordura saturada (31g de gordura saturada/100g de dieta hiperlipídica, HL) por cerca de 5 semanas, sendo ambas as dietas com acesso ad libitum. O peso dos filhotes desde o nascimento foi acompanhado semanalmente até o 1º mês de vida, passando a ser pesado mensalmente até a eutanásia. O comprimento naso-anal foi medido no DPN30 e no DPN60. Foram coletadas amostras de sangue nos DPN30, DPN60 e DPN90 para avaliação das concentrações sanguíneas de glicose, colesterol total e triglicérides em sangue total e das concentrações plasmáticas de insulina e leptina. Os animais foram submetidos aos testes comportamentais: Novelty-sepressed feeding test e Campo aberto. Após os acompanhamentos e avaliações comportamentais, os animais foram eutanasiados por decapitação e tiveram sangue, gordura, fígado, rins e músculo coletados. Os dados são apresentados como média ± desvio padrão. XVIII Resultados e conclusões: A RCIU diminuiu o peso ao nascer dos filhotes e esse efeito pode ser observado em outros momentos ao longo do estudo; nos machos, essa restrição reduziu o crescimento naso-anal e alterou a proporção de massa muscular e tecido adiposo após o consumo da dieta HL. Depois de 30 dias alimentando-se apenas com a dieta HL, os animais restritos apresentaram elevação da insulina plasmática. Machos e fêmeas do grupo RT apresentaram aumento da atividade locomotora. As fêmeas restritas também tiveram um menor número de episódios de auto-limpeza durante o teste comportamental, bem como apresentaram um maior consumo de ração HL ao longo do tempo em relação às fêmeas do grupo CT. Depois de passarem pelo desafio metabólico da exposição a dieta HL, os animais restritos apresentaram elevação da insulina plasmática, demonstrando que a RCIU pode influenciar sinalizadores metabólicos que participam da homeostase energética quando ocorre um desequilíbrio nutricional, além poder promover o aumento no consumo energético das fêmeas e aumentar a atividade locomotora de machos e fêmeas.
