Peptídeo mimético ao inibidor de PLA2 como uma nova ferramentapara o tratamento da asma

Resumo: | Allergic asthma is an inflammatory disease of global prevalence, characterized by symptoms as shortness of breath, wheezing and chest tightness, arising from a complex interaction between chronic inflammation and disordered airway function. Allergic asthma includes an organism sensitization by an antigen, which leads to respiratory parameters alterations, bronchiolar constriction, elevated production of specific IgE antibodies and elevated levels of eosinophils and phospholipase A2 enzymes (PLA2). Therefore, in this study, we investigated whether a peptide that mimics PLA2 inhibitor could mimetize its immunomodulatory effect, governing the inflammatory process in the respiratory tract and serve as a new therapeutic estrategy. These peptide (PF7), a bacteriophage (fF7) and dexamethasone (DEXA) were administered as tretaments to male Balb/C mice via intranasal, after ovalbumin sensitization and antigenic challenges. After 48 hours of the last challenge and treatment, mice respiratory electrophysiological parameters were analyzed, the bronchoalveolar lavage fluid (BAL), serum and lungs were collected for further analysis. Asthmatic mice that received PF7 had significant reductions in number of BAL eosinophils and serum IgE levels, indicating an attenuation of the inflammatory process. Electrophysiological parameters presented a significant improvement in pulmonary airflow. It also was observed by ATR-FTIR analysis the presence of amide I and amide II as potential biomarkers of allergic asthma. Our results demonstrated that treatment with peptide (PF7) attenuates the development of airway inflammation in ovalbumin-induced asthma, supporting the conclusion that strategies using peptides might be developed further as an attractive therapeutic approach in asthma treatment. |

Na asma alérgica inclui-se a sensibilização do organismo por um antígeno, que ocasionam alterações nos parâmetros respiratórios, constrição bronquiolar, produção elevada de anticorpos IgE específicos e níveis elevados de eosinófilos e fosfolipase A2 (PLA2). Em virtude disso, neste estudo investigamos se um peptídeo que mimetiza o inibidor da PLA2 poderia imitar seu efeito imunomodulador, governando o processo inflamatório no trato respiratório e servir como uma nova estratégia terapêutica. O peptídeo (PF7), um bacteriófago (fF7) e dexametasona (DEXA) foram administrados como tratamentos a camundongos Balb/C machos via intranasal, após sensibilização e desafios antigênicos por ovalbumina. Após 48 horas do último desafio e tratamento, os parâmetros eletrofisiológicos respiratórios dos camundongos foram analisados, o líquido de lavagem broncoalveolar (BAL), soro e pulmões foram coletados para análise posterior. Camundongos asmáticos tratados com peptídeo (PF7) tiveram reduções significativas no número de eosinófilos no BAL e níveis de IgE no soro, indicando atenuação do processo inflamatório. Os parâmetros eletrofisiológicos apresentaram melhora no fluxo de nos pulmões. Também foi possível observar na análise ATR-FTIR a presença de amida I e amida II, como potenciais biomarcadores de asma alérgica.
