Os CDs foram preparados a partir de rea??es de desidrata??o e oxida??o ?cida do biopol?mero celulose. A metodologia utilizada foi escolhida devido ? possibilidade de obten??o de nanopart?culas com alta concentra??o de grupos funcionais oxigenados, que ser?o ?teis para o liga??o do pept?deo antimicrobiano ecPis-4s nanopart?culas. A funcionaliza??o dos CDs foi realizada atrav?s da rea??o dos grupos carbonila e/ou hidroxila com o grupo amino ou am?nio do pept?deo por meio da ativa??o destes grupos usando 1-etil-3-(3-dimetilaminopropil) carbodiimida (EDAC) e acetato de etil-2-ciano-2-(hidroxiimino)(Oxyma pure?). Os materiais obtidos foram caracterizados com diferentes t?cnicas, incluindo Resson?ncia Magn?tica Nuclear de hidrog?nio (RMN ?H), espectroscopia na regi?o do infravermelho com transformada de Fourier (FTIR- do ingl?s Fourier Transform Infrared Spectroscopy) e potencial zeta. Ap?s a funcionaliza??o com o pept?deo, o potencial zeta das nanopart?culas de CDs passou de -50,5 mV para -23,4 mV, sugerindo o consumo de alguns grupos funcionais negativamente carregados, como os carbox?licos, em pH inferior a 6. Nos espectros no infravermelho e de RMN ?H dos CDs funcionalizados, ap?s purifica??o, p?de-se observar bandas caracter?sticas de ambos os compostos. Destaca-se, por exemplo, as absor??es em 1360 e 1050 cm-?, a primeira observada somente no espectro do CDs e a segunda bem definida no espectro do pept?deo. Em adi??o, observa-se a presen?a do grupo funcional amida por meio das absor??es em 1638 e 1560 cm-?. Al?m dessas observa??es, a funcionaliza??o pode ser indiretamente relacionada ao estreitamento da absor??o em 3340 cm-?(? O-H), o que sugere uma diminui??o das liga??es de hidrog?nio na amostra de CDs ap?s a rea??o. Se, por um lado, as nanoestruturas funcionalizadas n?o apresentaram atividade antioxidante, por outro, os resultados da atividade antimicrobiana in vitro, mostraram que as nanopart?culas funcionalizadas s?o capazes de inibir o crescimento bacteriano similarmente ao ecPis-4s para Staphylococcus aureus ( S. aureus), e Escherichia coli (E. coli), mostrando que elas podem inibir tanto o crescimento de bact?rias Gram-positivas quanto Gram?negativas. Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2021.Carbon dots (CDs) were functionalized with the ecPis-4s peptide, with the objective of obtaining a (bio)nanomaterial with antioxidant and antibacterial properties and future applications related with peptide-cell membrane interactions, such as biomarkers. The CDs were prepared from acid dehydration and oxidation reactions of the cellulose biopolymer. This methodology was chosen because, it allows the obtainment of allows the obtainment of nanoparticles with a high concentration of oxygenated functional groups, which are be useful for the binding of the antimicrobial peptide ecPis-4s on the nanoparticle. The functionalization of the CDs was performed through the reaction of the carboxyl and hidroxil groups (CDs) with the amino group of the peptide through the activation of carboxyl and/or OH groups using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDAC) and ethyl-2- cyano-2- (hydroxyimino) acetate (Oxyme Pure?). The materials obtained were characterized through different techniques, including proton Nuclear Magnetic Resonance of hydrogen (?H NMR), Fourier Transform Infrared Spectroscopy (FTIR) and zeta potential. After the functionalization with the peptide, the zeta potential of the CD nanoparticles changed from - 50.5 mV to -23.4 mV, suggesting the consumption of some negatively charged functional groups, such as carboxy, at a pH below 6. In the FTIR and ?H NMR spectra of the functionalized CDs, after purification, characteristic bands of both compounds were observed. For example, the absorptions at 1360 and 1050 cm-?, the former observed only in the spectrum of the CDs and the latter well-defined in the spectrum of the peptide. The presence of the amide functional group is observed through absorptions in 1638 and 1560 cm-?. In addition to these observations, the functionalization can be also indirectly related to the narrowing of absorption band around by 3340 cm-?(? O-H), which suggests a decrease in hydrogen bonds in the sample of CDs after the reaction. If, on the one hand, the functionalized nanostructures did not show antioxidant activity, on the other, the results of the antimicrobial activity, in vitro, showed that the functionalized nanoparticles are capable of inhibiting bacterial growth similary to ecPis-4s for Staphylococcus aureus (S. aureus), and Escherichia coli (E. coli), showing that they can inhibit growth with both Gram-positive and Gram-negative bacteria.
