Diversidade de fungos em solos de florestas plantadas de eucalipto

Resumo: The changes in the environment and the consequent reduction on the diversity under intensive cultural practices have taken the people to develop plans of conservation and sustainable use of land and forests management. The objective of this work was to study fungal diversity in commercial plantations of eucalypt, with different ages and management, in the South of Angola, East of Minas Gerais State in Brazil and three regions of Portugal. Samples were collected from January to December 2009. The ectomycorrhizal fungi diversity (ECMF) in the East of Minas Gerais State was evaluated by morphological and molecular characterization, and the arbuscular mycorrhizal fungi (AMF) occurrence was investigated by using spores. The total fungal diversity in the three counties was performed by molecular analysis of 18S rDNA of soil samples. All evaluated plants presented ectomycorrhizae, in all the times and areas in the East of Minas Gerais State. The highest colonization was settling in the oldest plants and in the second period time of sampling, suggesting the importance of ectomycorrhizal association in stress conditions, like in low temperature and water availability. At least nine different morphotypes of ectomycorrhizae were characterized. Basidiomes of Scleroderma and Pisolithus were found. The arbuscular mycorrhizal fungi Glomus, Acaulospora and Scutellosporawere observed under eucalypt plantations, and the first two species occurred in all evaluated soil. The morphological characterization of the spores associated to the molecular analysis of the 18S rDNA showed to have high similarities in the composition of the FMA community among soils of eucalypt plantations from Angola, Brazil and Portugal. The analysis of fungal composition using 18S rDNA showed considerable diversity in all three soils, and Brazilian’s presented greater index of species diversity. The results of this work show that it has great diversity of fungi in commercial plantations of eucalypt and the use of classic morphological and molecular techniques allows to better access and understanding the fungal diversity in forest environments.

Este trabalho teve como objetivo estudar a diversidade de fungos em plantios comerciais de eucalipto, com diferentes idades e manejos, naregião sul de Angola, região leste de Minas Gerais no Brasil e em três regiões distintas de Portugal. As coletas foram realizadas de janeiro a dezembro de 2009. A diversidade de fungos ectomicorrízicos (FECM) na região leste de Minas Gerais – Brasil foi avaliada pela caracterização morfológica das ectomicorrizas, e pela caracterização molecular das mesmas. A ocorrência de fungos micorrízicos arbusculares (FMA) foi avaliada pela densidade e caracterização morfológica e molecular dos esporos presentes nos solos. Foi também avaliada a diversidade de fungos totais do solo a partir da análise molecular do rDNA 18S em amostras de solo sob eucalipto nos três países. Observou-se a presença de ectomicorrizas em todas as plantas avaliadas, em todas as épocas e áreas com eucalipto de diferentes idades e manejo, na região leste de Minas Gerais - Brasil, havendo maior taxa de colonização nas plantas mais velhas e na segunda época de coleta, quando as condições climáticas eram de baixa precipitação e umidade relativa. A caracterização morfológica das ectomicorrizas mostrou a presença de pelo menos nove morfotipos. Foram encontrados basidiocarpos dos gêneros Scleroderma xi e Pisolithus, com predominância do primeiro. Foram identificados FMA dos gêneros Glomus, Acaulospora e Scutellospora a partir dos esporos dos solos, tendo os dois primeiros ocorrido em todos os solos avaliados. A caracterização morfológica dos esporos associada à análise molecular do gene 18S do rDNA dos esporos mostrou haver semelhanças na composição da comunidade de FMA dos solos da Angola, Brasil e Portugal. A análise molecular do DNA, a partir da extração direta do solo, mostrou a existência de uma diversidade considerável na composição das comunidades de fungos nos solos de florestas plantadas de eucalipto dos três países estudados, sendo que o solo do Brasil foi o que apresentou maior índice de diversidade de espécies.
