Use of analytical methodologies for the determination of phenolic compounds in food in Brazil: advances and weaknesses

Resumo: | The importance of food plants for human health transcends the nutritional aspect, and can contribute to the prevention of diseases, due to the presence of bioactive substances, among which some phenolic compounds stand out. Thus, the present work aims to carry out an integrative literature review on the main analytical methodologies used in Brazil for the extraction, identification, quantification and evaluation of the antioxidant activity (AA) of total phenolics (TF) in foods, addressing their advances and weaknesses. It was observed that a universal protocol for studies of TF and its AA has not yet been established, as factors such as the origin of the sample are still preponderant for the choice of methods. The high cost is the main limiting factor for the wide use of more modern techniques such as lyophilization, despite other unconventional methodologies such as ultrasound (US) and microwave extraction are not becoming more popular. Maceration, solvent extraction, percolation and cold extraction will remain the most common extraction methods in Brazil. Although the use of chromatographic techniques, such as HPLC, coupled with powerful detectors are more accurate for quantification and identification, the high cost has also restricted the popularization of its use, with the Folin-Ciocalteau spectrophotometric technique remaining as the most widespread, due to its simplicity, reproducibility and convenience. As for the measurement of AA, it is common to use more than one methodology for its determination, in order to overcome the weaknesses of each method. |

Así, el presente trabajo tiene como objetivo realizar una revisión integradora de la literatura sobre las principales metodologías analíticas utilizadas en Brasil para la extracción, identificación, cuantificación y evaluación de la actividad antioxidante (AA) de los fenólicos totales (FT) en los alimentos, abordando sus avances y debilidades. Se observó que aún no se ha establecido un protocolo universal para estudios de FT y su AA, ya que factores como el origen de la muestra siguen siendo preponderantes para la elección de métodos. El alto costo es el principal factor limitante para el amplio uso de técnicas más modernas como la liofilización, a pesar de que otras metodologías no convencionales como el ultrasonido y la extracción por microondas se están volviendo más populares. La maceración y extracción por solventes seguirán siendo los métodos de extracción más comunes en Brasil. Si bien el uso de técnicas cromatográficas, como HPLC, junto con detectores potentes son más precisas para la cuantificación e identificación, el alto costo también ha restringido su popularización, quedando la técnica espectrofotométrica de Folin-Ciocalteau como la más extendida, debido a su sencillez, reproducibilidad y conveniencia. En cuanto a la medición de AA, es común utilizar más de una metodología para su determinación, con el fin de superar las debilidades de cada método.A importância das plantas alimentícias para a saúde humana transcende o aspecto nutricional, podendo contribuir para a prevenção de doenças, em função da presença de substâncias bioativas, dentre as quais se destacam alguns compostos fenólicos. Desta forma, o presente trabalho tem por objetivo fazer uma revisão integrativa da literatura sobre as principais metodologias analíticas utilizadas no Brasil para extração, identificação, quantificação e avaliação da atividade antioxidante (AA) de fenólicos totais (FT) em alimentos, abordando seus avanços e fragilidades. Observou-se que ainda não foi estabelecido um protocolo universal para estudos de FT e de sua AA, pois fatores como origem da amostra ainda são preponderantes para a escolha dos métodos. O elevado custo é o principal fator limitador para a ampla utilização de técnicas mais modernas como a liofilização, apesar de outras metodologias não-convencionais como extração por ultrassom (US) e micro-ondas estarem se popularizando.
