Enterococcus faecium EF137V: a new strategic source for the control of human and animal health against Campylobacter species

Resumo: The probiotic is related to a food supplement, while the live biotherapeutic products - LBPs are considered biological medicines, both contain living microorganisms with active substances that benefit the host. Enterococcus faecium has many interesting and functional properties for human and animal health, including the antimicrobial capacity, which helps in food poisoning diseases such as Campylobacteriosis. Campylobacter jejuni and C. coli species have been described in this context. Therefore, due to the need to develop alternatives to ensure the safety of products consumed by the population, this work proposes a complementary strategy to reduce or eliminate Campylobacter through the use of Enterococcus faecium EF137V, isolated from artisanal “Coalho” cheese from Pernambuco-Brazil. The analyzes were performed in vitro. Given the adverse conditions, such as acid pH and the presence of bile salts, in which EF137V was exposed, it was found that the bacteria showed resistance to gastrointestinal conditions, adherence to epithelial cells, antimicrobial activity by its metabolic compounds against reference strains of C. jejuni and C. coli, as well as antagonistic activity, which suggests that both species will not be able to grow disorderly in the host's intestinal microbiota, thus being able to control these microorganisms in the bacterial community in broilers and preventing new infections in humans. It can be concluded that the EF137V is a promising PVB and probiotic, presenting great capacity for biotechnology applications.

Enterococcus faecium tiene muchas propiedades interesantes y funcionales para la salud humana y animal, incluida la capacidad antimicrobiana, que ayuda en enfermedades de intoxicación alimentaria como la campilobacteriosis. En este contexto se han descrito especies de Campylobacter jejuni y C. coli. Por lo tanto, ante la necesidad de desarrollar alternativas para asegurar la inocuidad de los productos consumidos por la población, este trabajo propone una estrategia complementaria para reducir o eliminar Campylobacter mediante el uso de Enterococcus faecium EF137V, aislado de queso “Coalho” artesanal de Pernambuco-Brasil. Los análisis se realizaron in vitro. Dadas las condiciones adversas, como pH ácido y presencia de sales biliares, en las que se expuso EF137V, se encontró que la bacteria presentaba resistencia a afecciones gastrointestinales, adherencia a células epiteliales, actividad antimicrobiana por sus compuestos metabólicos frente a cepas de referencia de C. jejuni y C. coli, así como actividad antagónica, lo que sugiere que ambas especies no podrán crecer desordenadamente en la microbiota intestinal del hospedador, pudiendo así controlar estos microorganismos en la comunidad bacteriana en pollos de engorde y prevenir nuevas infecciones en humanos. Se puede concluir que el EF137V es un PVB y probiótico prometedor, presentando gran capacidad para aplicaciones biotecnológicas.O probiótico está relacionado a um suplemento alimentar, enquanto que os produtos vivos bioterapêuticos - PVBs são considerados medicamentos biológicos, ambos contêm micro-organismos vivos com substâncias ativas e que beneficiam o hospedeiro. As espécies Campylobacter jejuni e C. coli, têm sido descritas neste contexto. Portanto, pela necessidade de desenvolver alternativas que permitam garantir a segurança dos produtos consumidos pela população, este trabalho propõe uma estratégia complementar para reduzir ou eliminar o Campylobacter por meio do uso de Enterococcus faecium EF137V, isolado de queijo “Coalho” artesanal de Pernambuco-Brasil. As análises foram realizadas in vitro. Dadas as condições adversas, como pH ácido e presença de sais biliares, nas quais EF137V foi exposto, verificou-se que a bactéria apresentou resistência a condições gastrointestinais, aderência às células epiteliais, atividade antimicrobiana pelos seus compostos metabólicos contra cepas de referência de C. jejuni e C. coli, bem como atividade antagonista, o que sugere que ambas as espécies não serão capazes de crescer desordenadamente na microbiota intestinal do hospedeiro, podendo assim controlar a esses microrganismos na comunidade bacteriana em frangos de corte e evitando novas infecções em humanos.
