Email: msouza@ufv.br The aim of this research was to verify the therapeutic effects of platelet-rich plasma (PRP) in the treatment of tendinopathy of the superficial digital flexor tendon as assessed by histological and morphometric evaluation, as well as to evaluate early changes in the expression of factor VIII by immunohistochemical analysis during the inflammatory and proliferative phases of healing process of PRP treated tendons compared to placebo. Five days after induction of tendinopathy by collagenase, the forelimbs of six horses were treated with intralesional PRP or 0.9% NaCl. Tendon biopsy was performed at 3 or 16 days after treatments. In both phases and with both treatments, the lesion area was filled with granulation tissue containing scattered collagen fibers with loss of parallelism, fibroblasts with varied morphology, neovascularization and hypertrophied endotenon. These characteristics were more evident during the proliferative phase and no differences were observed between treatments. A negative correlation between collagen fibers and moderately plump fibroblasts was observed. Immunohistochemistry staining was positive for factor VIII, without any differences between the treatments and tendon healing phases.

Immunohistochemistry staining was positive for factor VIII, without any differences between the treatments and tendon healing phases.O objetivo desse estudo foi verificar o efeito terapêutico do plasma rico em plaquetas (PRP) no tratamento de tendinopatia do tendão do músculo flexor digital superficial mediante realização de estudos histológico e morfométrico, bem como avaliar alterações iniciais na expressão do fator VIII por imunoistoquímica durante as fases inflamatória e proliferativa do processo de reparação do tendão tratado com PRP e com placebo. Para isso, cinco dias após indução de tendinopatia por colagenase, os membros torácicos de seis equinos foram tratados por via intralesional com PRP ou NaCl a 0,9%. Biópsia tendínea foi realizada 3 ou 16 dias após os tratamentos. Em ambas as fases e ambos os tratamentos a área da lesão estava preenchida por tecido de granulação contendo fibras colágenas esparsas, com perda do paralelismo e fibroblastos com morfologia variada, neovascularização e hipertrofia do endotendão. Essas características foram mais evidentes durante a fase proliferativa, não havendo diferença entre os tratamentos. Correlação negativa foi observada entre a presença de fibras colágenas e fibroblastos moderadamente arredondados.
