A transformation protocol using mannose as selective agent was evaluated for genetic transformation of the rootstock Swingle citrumelo. Two independent experiments were carried out to define the smallest mannose concentration capable to inhibit the multiplication of adventitious buds. Based on these results, 5.0 mM of mannose was chosen for Swingle citrumelo genetic transformation. An 11% transformation efficiency was obtained using the Agrobacterium tumefaciens binary strain EHA105. The results described in this study showed that, despite the relatively low transformation frequency, the gene pmi is a good alternative to the most commonly used selection scheme for citrus transformation, based on gene nptII for kanamycin resistance. |
