Elizabeth TeodorovDissertação (mestrado) - Universidade Federal do ABC, Programa de Pós-Graduação em Biotecnociência, Santo André, 2018.O uso de farmacos veterinarios para tratamento de doencas das mais diferentes variabilidades vem crescendo exponencialmente no mundo. A ivermectina tem amplo uso para tratamento de ectoparasitas e helmintos e na aquicultura como profilaxia aos peixes, porem entra no corpo d'agua e afeta negativamente os organismos. Esse estudo teve como objetivo avaliar aspectos toxicologicos e teratogenicos em estagio embriolarval de Zebrafish (Danio rerio), submetido a decorionizacao ou nao e exposto a diferentes concentracoes de ivermectina diluidos em solventes diferentes sendo agua destilada e DMSO. Os resultados demonstraram que, na curva dose-resposta, concentracoes de ivermectina acima de 120 ¿Êg/L foram letais aos individuos. Ainda independente do diluente, observou-se maior taxa de mortalidade em concentracoes mais baixas de ivermectina ao passo que nas concentracoes mais altas foi observado intenso edema. Em relacao aos resultados teratogenicos observou-se que as principais malformacoes encontradas em individuos expostos a ivermectina incluiram edema cardiaco e malformacoes de cabeca e cauda. Como o corion e um envoltorio que protege o animal em sua fase embrionaria de agentes quimicos conforme sua estrutura quimica realizou-se o experimento com decorionizacao (retirada enzimatica do corion) e os resultados revelaram que as larvas expostas a ivermectina apresentaram prejuizos similares aqueles que nao sofreram a decorionizacao. Em relacao a atividade locomotora das larvas expostas as concentracoes mais altas de ivermectina diluida em DMSO os dados mostram prejuizo deste parametro, sendo que nas concentracoes de 60 ¿Êg/L e 120 ¿Êg/L as larvas permaneceram imoveis, possivelmente por uma acao GABAergica. Conclui-se, portanto que a ivermectina em altas concentracoes apresentou ambos os efeitos toxico e teratogenico, evidenciado particularmente pelas malformacoes encontradas e que sao incompativeis com a vida; esses achados reforcam a preocupacao quanto a dose correta, prescrita por medicos veterinarios quando do tratamento anti-parasitario de seus pacientes.The use of veterinary drugs to treat diseases of different variability has been growing exponentially in the world. Ivermectin has wide use for the treatment of ectoparasites and helminths and in aquaculture is used as a prophylaxis for fish, but itLs enters in the body of water and caude negatively affects in organisms. The objective of this study was to evaluate the toxicological and teratogenic aspects of embryo and larvae Zebrafish (Danio rerio), submitted or not to decorionization and exposed to many concentrations of ivermectin diluted in different solvents, such as distilled water and DMSO. The results demonstrated that, in the dose-response curve experiment, concentrations of ivermectin above 120 ¿Êg/L were lethal. Also, independent of the diluent, a higher mortality was observed at lower concentrations of ivermectin, whereas in the higher concentrations an intense edema was observed. Regarding the teratogenic results, it was observed that the main malformations found in subjects exposed to ivermectin included cardiac edema and head and tail malformations. As the chorion is a wrap that protects the fish in its embryonic stage of chemical agents the experiment was carried out with decorionization (enzymatic removal of the chorion) and the results revealed that the larvae exposed to ivermectin presented damages similar to those that did not undergo decorionization. In relation to the locomotor activity of the larvae exposed to the highest concentrations of ivermectin diluted in DMSO, the data show a loss of this parameter, and in the concentrations of 60 ¿Êg/L and 120 ¿Êg/L the larvae remained immobile, possibly due to a GABAergic action.
