Resumo: | RESUMO CAPÍTULO I The araticum (Annona crassiflora Mart.) is a typical brazilian plant found in cerrados of Brazil. This plant contains acetogenins that presents cytotoxic, antitumorigenic, antiparasitic and antimicrobial properties. Its leaves, barks, fruits and seeds are used by the population as therapeutic medicine to treat several diseases as diarrhoea, rheumatism and syphilis. In the present study we have evaluated the mutagenic activity of the crude ethanolic extract of leaves of araticum though quantification of micronuclei induced in mice bone marrow. Doses of 10 mg/kg (5% of LD50), 20 mg/kg (10% of LD50), 50 mg/kg (25% of LD50), 100 mg/kg (50% of LD50) and 160 mg/kg (80% of LD50) were applied i.p. in mice Mus musculus, in groups of 5 (five) animals for each dose. The cytological preparations were made according to Heddle`s methodology. For all the applied doses, frequency of micronucleated polychromatic erythrocytes (MNPCE) was evaluated after 24, 48 and 72 h of treatment. The cytotoxicity was evaluated by ratio the between numbers of polychromatic and normochromatic erythrocytes (PCE/NCE). Ours results indicated an absence of significantly increased of micronuclei for all the doses tested (p>0,01). Thus, we concluded that the phytoterapic araticum did not present mutagenic activity under our experimental conditions. Suas folhas, cascas, frutos e sementes são utilizadas pela população para tratar várias doenças como diarréia, reumatismo e sífilis. No presente estudo, foi avaliada a atividade mutagênica do extrato bruto etanólico de folhas de araticum pelo teste do micronúcleo na medula óssea de camundongos. Doses de 10 mg/kg (5% da DL50), 20 mg/kg (10% da DL50), 50 mg/kg (25% da DL50), 100 mg/kg (50% da DL50) e 160 mg/kg (80% da DL50) foram aplicadas, intraperitonealmente, em camundongos Mus musculus, em grupos de 5 animais para cada dose. As preparações citológicas foram realizadas em conformidade com a metodologia de Heddle. Para todas as doses aplicadas, a freqüência de eritrócitos policromáticos micronucleados (EPCMN) foi avaliada nos tempos de 24, 48 e 72 h. A citotoxicidade foi avaliada pela relação entre a freqüência de eritrócitos policromáticos e normocromáticos (EPC/ENC). Os resultados obtidos mostraram que o extrato bruto etanólico de folhas, para todas as doses testadas, não aumentou significativamente (p>0,01) a freqüência de eritrócitos policromáticos micronucleados. Dessa maneira, pôde-se concluir que o fitorápico araticum não exibiu atividade mutagênica. Entretanto, a atividade tóxica foi verificada pela DL50 e pela relação EPC/ENC. Doses do extrato de folhas (10 mg/kg, 20 mg/kg, 50 mg/kg, 100 mg/kg e 160 mg/kg) e MMC (4,0 mg/kg) foram co-administradas, via intraperitoneal, em camundongos Mus musculus, em grupos de 5 (cinco) animais para cada dose. As preparações citológicas foram realizadas conforme metodologia de Heddle. Para todas as doses aplicadas, a freqüência de eritrócitos policromáticos micronucleados (EPCMN) foi analisada 36 horas após tratamento. Os resultados obtidos mostraram que o extrato de folhas nas doses de 20 mg/kg, 50 mg/kg e 100 mg/kg inibiram significantemente (P<0,01) a freqüência de micronúcleos (MN) induzidos pela MMC.
