Passeriformes versus n?o Passeriformes: compara??o filogen?tica atrav?s da atividade da enzima lactato desidrogenase plasm?tica

Resumo: This study quantified the activity of lactate dehydrogenase (LDH) from passerines (Serinus canarius) (n = 32) and non passerines (Melopsittacus undulatus) (n = 32), from the State of Rio de Janeiro, Brazil, to establish differences between the plasm activities of LDH, using this enzyme as a phylogenetic character. The blood was collected through the jugular vein puncture with syringe BD Ultra Fine II Short volume of 0.3 ml to Belgian canaries and l ml in Budgerigars. Before being released the animals were weighed on digital scale for accuracy GEHAKA model BG 200. After centrifugation of samples, heparinized plasm was separated and frozen at 4 ? C, and biochemically examined the following day, through biochemical kits for LDH UV - PP's Gold Analysis (kinetic method) and read in spectrophotometer model BIOPLUS 2000. The data of mass (kg), BMR (Kcal / day), BMRS (Kcal / kg / day) and quantification of enzyme LDH (U / L) were subjected to calculation of mean and standard deviation for Excel 2003. Then the means were compared using the test (t) Student at 5% level of probability, using the statistical program of Statistic 7.0. The results were plotted, relating the activities of LDH to the masses, the LDH to BMRs and the LDH to BMRSs. Passerines showed the mass of 0019 ? 0002, BMR of 6.67 ? 0.58, BMRS of 1949.84 ? 55.76 and LDH of 1560.16 ? 432.05. In non passerines, mass of 0036 ? 0004, BMR of 6.42 ? 0.57, BMRS of 1009.95 ? 29.27 and LDH of 123 ? 56. All data are normal, parametric, and the t test was possible to verify that there is significant difference between masses, BMRSs and LDHs in passerines and non passerines. There is no significant difference in BMR between birds. Passerines and non passerines minors have lower BMR and BMRS greater. The activity of LDH is mass-dependent within the same species, but not when compared passerines and non passerines you can see that the passerines, although smaller, are phylogenetically more anaerobic.

O sangue foi coletado atrav?s de pun??o na veia jugular, com seringa BD Ultra Fine II curta de volume de 0,3ml nos can?rios belga e 1 ml nos periquitos australianos. Antes de serem liberados os animais foram pesados em balan?a digital de precis?o GEHAKA modelo BG 200. Ap?s centrifuga??o das amostras, o plasma heparinizado foi separado e congelado a 4? C, sendo analisado bioquimicamente no dia seguinte, atrav?s de kits de bioqu?mica de LDH UV PP da Gold Analisa (m?todo cin?tico) e leitura em espectrofot?metro modelo BIOPLUS 2000. Os dados de massa (Kg), TMB (Kcal/dia), TMBE (Kcal/Kg/dia) e quantifica??o das enzimas de LDH (U/L) foram submetidos ao c?lculo da m?dia e desvio padr?o pelo Excel 2003. Em seguida as m?dias foram comparadas atrav?s do teste (t) de Student ao n?vel de 5% de probabilidade, com aux?lio do programa de estat?stica Statistic 7,0. Os resultados foram plotados, relacionando as atividades de LDH ?s massas, LDH ?s TMBs e LDH ?s TMBEs dos passeriformes e n?o passeriformes. Os passeriformes apresentaram massa de 0,019 ? 0,002, TMB de 6,67 ? 0,58, TMBE de 346,74 ? 9,92 e LDH, 1560,16 ? 432,05. Em n?o passeriformes, massa de 0,036 ? 0,004, TMB de 6,42 ? 0,57, TMBE de 179,60 ? 5,21 e LDH, 123 ? 56. Todos os dados s?o normais e param?tricos, e ao teste t foi poss?vel verificar que existe diferen?a significativa entre massas, TMBEs e LDHs de passeriformes e n?o passeriformes. N?o existe diferen?a significativa em TMB entre as aves. Passeriformes e n?o passeriformes menores apresentam TMB menor e TMBE maior. A atividade da LDH ? massa-dependente dentro de uma mesma esp?cie, por?m quando comparamos passeriformes e n?o passeriformes, ? poss?vel verificar que os passeriformes, mesmo sendo menores, s?o filogeneticamente mais anaer?bicos.Made available in DSpace on 2016-04-26T19:00:03Z (GMT).
