Resumo: The urban pigeon (Columba livia) is a bird frequently observed in urban centers, living in close contact with humans, being a possible disseminator of pathogens in the environment. Among these, special attention should be given to the species of Cryptosporidium, since the pigeon can harbor and eliminate the oocysts through the faeces. To date, three species are considered valid parasitizing birds (Cryptosporidium meleagridis, Cryptosporidium baileyi and Cryptosporidium galli) and 13 genotypes as possible agents of avian cryptosporidiosis. As the first species, besides parasitizing the birds class, has been diagnosed in several mammalian hosts, also zoonotic. Due to the lack of studies on the diagnosis of Cryptosporidium spp. parasitizing pigeons in Brazil and with few reports in the world, the present work had as objectives: to diagnose microscopically the presence of oocysts of Cryptosporidium spp. from pigeons faecal samples captured in three areas in the Rio de Janeiro city; to characterize genotypes and / or genotypes and / or subtypes obtained from those positive samples for Cryptosporidium spp. and perform sequencing and phylogenetic analyzes by comparing the sequences obtained with those sequences deposited in GenBank. A total of 387 pigeons fecal samples were collected in three administrative regions: Campo Grande - denominated A (98 samples), Bangu - denominated B (187 samples) and Madureira - denominated C (102 samples). Initially the Cryptosporidium spp. oocysts were investigated. For this, the faeces were processed through the technique of centrifugation and flotation in saturated sugar solution and evaluated by light microscopy. In the microscopic diagnosis, 81 (20.93%) samples were positive for the presence of Cryptosporidium spp. oocysts, distributed: 28.82% in area A, 22.99% in area B and 9.80% in area C. In these, DNA extraction was performed, followed by amplification and sequencing for the 18S target. Of these 81, 53 samples amplified in Nested-PCR. Subsequently, these samples were purified and sent for sequencing, followed by phylogenetic analyzes. In this study, two species were diagnosed: Cryptosporidium meleagridis in 44 samples (91.67%) and Cryptosporidum baileyi in four samples (8.33%), both of which were more closely related to intestinal species. Sequences of seven samples containing different nucleotides of Cryptosporidum were deposited in Genbank. So far, the report of this pathogen in pigeons had only occurred in four countries in the world: Turkey, Iran, China and Thailand. The data obtained in this study reflect public health concern, and C. meleagridis may present zoonotic potential. This study is the first Brazilian registry to have the host pigeon (Columba livia) infected by Cryptosporidium, contributing to trace the molecular epidemiology of this etiological agent in the country

Devido a aus?ncia de estudos sobre o diagn?stico de Cryptosporidium spp. parasitando pombos no Brasil e com escassos relatos no mundo, o presente trabalho teve como objetivos: diagnosticar microscopicamente a presen?a de oocistos de Cryptosporidium spp. provenientes de amostras fecais de pombos capturados em tr?s ?reas no munic?pio do Rio de Janeiro; caracterizar genotipicamente esp?cies e/ou gen?tipos e/ou subtipos obtidos daquelas amostras positivas para Cryptosporidium spp. e realizar o sequenciamento e an?lises filogen?ticas, comparando as sequencias obtidas com aquelas sequ?ncias depositadas no GenBank. Foram coletados um total de 387 amostras fecais de pombos, em tr?s regi?es administrativas: Campo Grande ? denominada de A (98 amostras), Bangu ? denominada de B (187 amostras) e Madureira ? denominada de C (102 amostras). Inicialmente foi realizada a pesquisa de oocistos Cryptosporidium spp., para tal, as fezes foram processadas atrav?s da t?cnica de centrifuga??o e flutua??o em solu??o saturada de a??car e avaliadas por microscopia ?ptica. No diagn?stico microsc?pico, 81 (20,93%) amostras apresentaram positividade para a presen?a de oocistos de Cryptosporidium spp., distribu?dos: 28,82% na ?rea A, 22,99% na ?rea B e 9,80% na ?rea C. Nestas, foi realizada a extra??o de DNA, seguida de amplifica??o e sequenciamento para o alvo 18S. Sendo que destas 81, 53 amostras amplificaram na Nested-PCR. Posteriormente, estas amostras foram purificadas e encaminhadas para o sequenciamento, seguido de an?lises filogen?ticas. Neste estudo, foram diagnosticadas duas esp?cies: Cryptosporidium meleagridis em 44 amostras (91,67%) e Cryptosporidum baileyi em quatro amostras (8,33%), ambas se apresentando mais estreitamente relacionadas ?s esp?cies intestinais. As sequ?ncias de sete amostras contendo nucleot?deos diferentes de Cryptosporidum foram depositadas no Genbank. At? o momento o relato deste pat?geno em pombos s? havia ocorrido em quatro pa?ses no mundo: Turquia, Ir?, China e Tail?ndia. Os dados obtidos neste estudo refletem preocupa??o em sa?de p?blica, sendo que a C. meleagridis pode apresentar potencial zoon?tico. description The urban pigeon (Columba livia) is a bird frequently observed in urban centers, living in close contact with humans, being a possible disseminator of pathogens in the environment. Among these, special attention should be given to the species of Cryptosporidium, since the pigeon can harbor and eliminate the oocysts through the faeces. To date, three species are considered valid parasitizing birds (Cryptosporidium meleagridis, Cryptosporidium baileyi and Cryptosporidium galli) and 13 genotypes as possible agents of avian cryptosporidiosis. As the first species, besides parasitizing the birds class, has been diagnosed in several mammalian hosts, also zoonotic. Due to the lack of studies on the diagnosis of Cryptosporidium spp. parasitizing pigeons in Brazil and with few reports in the world, the present work had as objectives: to diagnose microscopically the presence of oocysts of Cryptosporidium spp. from pigeons faecal samples captured in three areas in the Rio de Janeiro city; to characterize genotypes and / or genotypes and / or subtypes obtained from those positive samples for Cryptosporidium spp. and perform sequencing and phylogenetic analyzes by comparing the sequences obtained with those sequences deposited in GenBank. A total of 387 pigeons fecal samples were collected in three administrative regions: Campo Grande - denominated A (98 samples), Bangu - denominated B (187 samples) and Madureira - denominated C (102 samples). Initially the Cryptosporidium spp. oocysts were investigated. For this, the faeces were processed through the technique of centrifugation and flotation in saturated sugar solution and evaluated by light microscopy. In the microscopic diagnosis, 81 (20.93%) samples were positive for the presence of Cryptosporidium spp. oocysts, distributed: 28.82% in area A, 22.99% in area B and 9.80% in area C. In these, DNA extraction was performed, followed by amplification and sequencing for the 18S target. Of these 81, 53 samples amplified in Nested-PCR. Subsequently, these samples were purified and sent for sequencing, followed by phylogenetic analyzes. In this study, two species were diagnosed: Cryptosporidium meleagridis in 44 samples (91.67%) and Cryptosporidum baileyi in four samples (8.33%), both of which were more closely related to intestinal species. Sequences of seven samples containing different nucleotides of Cryptosporidum were deposited in Genbank. So far, the report of this pathogen in pigeons had only occurred in four countries in the world: Turkey, Iran, China and Thailand. The data obtained in this study reflect public health concern, and C. meleagridis may present zoonotic potential. This study is the first Brazilian registry to have the host pigeon (Columba livia) infected by Cryptosporidium, contributing to trace the molecular epidemiology of this etiological agent in the country
