Resumo: Endocrine disrupters are substances that impair the normal functioning of the endocrine system of animals, including humans. They include nonylphenol (NP), a degradation product of alkylphenol ethoxylate (APE), widely used nonionic surfactant. The development of appropriate method for determining these xenobiotics becomes essential. The goal of this study is to develop an analytical procedure used high performance liquid chromatography with ultraviolet detection (HPLC-UV) for the determination of nonylphenol in surface waters. In the procedure of the concentration and also extraction of nonylphenol was achieved using solid phase extraction (cartridge C18, 500 mg). The separation was performed using the Zorbax Eclipse Plus C18 chromatographic column (4.6 mm ID x 100 mm, 3.5 ?m), with a 15 minute running time, with a mobile phase of acetonitrile and ultra pure water isocratic mode with a flow rate of 1 mL min-1 and a wavelength of 225 nm. The method was validated for the parameters listed USEPA 8000D method. The recovery of nonylphenol was found in the range of 85% and the repeatability was adequate (relative standard deviation < 10%). The method detection and quantification limits were 0.18 and 1,15 ?g L-1 respectively. After validation of the method was applied in surface water samples collected in the hydrographic basin of the river Guandu, Rio de Janeiro, Brazil. The samples were collected in Itaguai, Serop?dica, Pirai, Paracambi, Santa Cruz (Rio de Janeiro), Queimados and Japeri. The occurrence of nonylphenol was observed in six of the seven collection points, where the concentration of nonylphenol ranged from values below the quantification limit to values of 2.32 ?g L-1, the latter found in the municipality of Paracambi. The result may suggest that the region under consideration may be impacted by the presence of nonylphenol, promoting damage to the environment and to humans. The test procedure proposed by SPE and HPLC-UV was efficient in determining nonylphenol within the watershed of the Guandu River region.

A separa??o foi feita utilizando-se uma coluna cromatogr?fica C18 Zorbax Eclipse Plus (4,6 mm ID x 100 mm, 3,5 ?m), com um tempo de corrida de 15 minutos, com a fase m?vel de acetonitrila e ?gua ultra pura em modo isocr?tico com uma vaz?o de 1 mL min-1, e comprimento de onda de 225 nm. O m?todo foi validado utilizando-se os par?metros de valida??o da USEPA (United States Environmental Protection Agency), metodologia 8000D. A taxa de recupera??o de nonilfenol ficou na faixa de 85 %, e a repetibilidade foi adequada (desvio padr?o relativo < 10 %). O limite de detec??o e quantifica??o do m?todo foram 0,2 e 1,15 ?g L-1 respectivamente. Ap?s a valida??o da metodologia, a mesma foi aplicada em amostras de ?gua coletadas na bacia hidrogr?fica do rio Guandu, Rio de Janeiro, Brasil. As amostras foram coletadas em Itagua?, Serop?dica, Pira?, Paracambi, Santa Cruz (Rio de Janeiro), Queimados e Japeri. Foi observada a ocorr?ncia do nonilfenol em seis dos sete pontos de coleta, nos quais a concentra??o do nonilfenol variou de valores abaixo do limite de quantifica??o at? valores de 2,32 ?g L-1, encontrado no mun?cipio de Paracambi. O resultado pode sugerir que a regi?o em estudo pode estar sendo impactada com a presen?a de nonilfenol, promovendo danos ao meio ambiente e ao homem. description Endocrine disrupters are substances that impair the normal functioning of the endocrine system of animals, including humans. They include nonylphenol (NP), a degradation product of alkylphenol ethoxylate (APE), widely used nonionic surfactant. The development of appropriate method for determining these xenobiotics becomes essential. The goal of this study is to develop an analytical procedure used high performance liquid chromatography with ultraviolet detection (HPLC-UV) for the determination of nonylphenol in surface waters. In the procedure of the concentration and also extraction of nonylphenol was achieved using solid phase extraction (cartridge C18, 500 mg). The separation was performed using the Zorbax Eclipse Plus C18 chromatographic column (4.6 mm ID x 100 mm, 3.5 ?m), with a 15 minute running time, with a mobile phase of acetonitrile and ultra pure water isocratic mode with a flow rate of 1 mL min-1 and a wavelength of 225 nm. The method was validated for the parameters listed USEPA 8000D method. The recovery of nonylphenol was found in the range of 85% and the repeatability was adequate (relative standard deviation < 10%). The method detection and quantification limits were 0.18 and 1,15 ?g L-1 respectively. After validation of the method was applied in surface water samples collected in the hydrographic basin of the river Guandu, Rio de Janeiro, Brazil. The samples were collected in Itaguai, Serop?dica, Pirai, Paracambi, Santa Cruz (Rio de Janeiro), Queimados and Japeri. The occurrence of nonylphenol was observed in six of the seven collection points, where the concentration of nonylphenol ranged from values below the quantification limit to values of 2.32 ?g L-1, the latter found in the municipality of Paracambi. The result may suggest that the region under consideration may be impacted by the presence of nonylphenol, promoting damage to the environment and to humans. The test procedure proposed by SPE and HPLC-UV was efficient in determining nonylphenol within the watershed of the Guandu River region.
