Resumo: | To evaluate the immune response of BALB/c mice immunized with the synthetic peptide anti-tick R. (B.) microplus SBm7462 by oral and nasal routes and to study the induction of AICD in memory T cells were used 80 BALB/c mice, divided in four experimental groups with 20 animals each (group I: oral immunization; group II: nasal immunization; group III: animals not immunized; and a group with animals givers of peritoneal macrophages. Two immunizations had been made with an interval of 21 days. At nine and 15 days after the first and the second immunization the collects were realized, separately for each group, and the T cells isolated had been cultivated with 5% CO2 at 37ºC, per 10 days in the presence of 1μg of SBm7462 and APCs. Later, the different agents, inductive (mithramycin and cycloheximide) or inhibiting (benzamide riboside and cyclosporin A) of apoptosis had been added, subdivided in distinct treatments. After the specific incubation period for each one of the treatments, it had been determined the percentage of cellular viability by the exclusion for Tripan Blue and the percentage of apoptotic cells with the dye Acridine Orange and by TUNEL technique. The specific nature of the T cells was confirmed by the increase in the number of cells after re-stimulation in vitro with SBm7462, which had held as memory T cells. The induction of AICD occurred by the inhibition of the expression of c-myc and by the increase of the expression of Fas and the induced AICD for the synthetic peptide SBm7462 was mediated by an independent way of the calcineurina and the kinetic of regulation of FasL. The mucosa routes, oral and nasal, can be considered alternative ways for the immunization with the synthetic peptide SBm7462 however they require a bigger number of doses to induce a response with high levels of immunoglobulins. |

(B.) microplus SBm7462 e estudar a indução da AICD dos linfócitos T de memória dos animais imunizados, utilizou-se 80 camundongos BALB/c, divididos em quatro grupos experimentais com 20 animais cada (grupo I: imunização por via oral; grupo II: imunização por via nasal; grupo III: animais não imunizados; e um grupo de animais doadores de macrófagos peritoneais). Foram feitas duas imunizações, com intervalo de 21 dias. Aos nove e 15 dias após a primeira e segunda imunização realizou-se as coletas, separadamente para cada grupo, e os linfócitos T isolados foram cultivados em estufa, 5% de CO2 a 37ºC, por 10 dias na presença de 1μg de SBm7462 e APCs. Posteriormente, adicionou-se os diferentes agentes indutores (mitramicina e CHX) ou inibidores (DBR e CsA) de apoptose, subdivididos em tratamentos distintos. Após o período de incubação específico para cada um dos tratamentos, determinou-se a porcentagem de viabilidade celular pela exclusão pelo Azul de Tripan e a porcentagem de células apoptóticas pela coloração com Laranja de Acridina e pela técnica de TUNEL. A natureza específica dos linfócitos T foi confirmada pelo aumento no número de células após re-estimulação in vitro com SBm7462, os quais se comportaram como linfócitos T de memória. Ocorreu indução da AICD por inibição da expressão do c-myc e por aumento da expressão de Fas e a AICD induzida pelo peptídeo sintético SBm7462 foi mediada por uma via independente da calcineurina e da cinética de regulação do FasL.
