Identification of altered microRNA target genes and pathways in cervical intraepithelial neoplasia

Resumo: MicroRNAs are single-stranded, non-protein-coding RNA molecules of about 18-25 nucleotides, which are capable of regulating expression at the post-transcriptional level. Its dysregulation has been widely studied in cancer, but little has been documented in pre-malignant lesions, such as Cervical Intraepithelial Neoplasms (CIN). In this context, this study aims to search the literature for microRNAs associated with NICs, as well as perform in silico prediction, in order to know the main genes and pathways that are regulated by them. For this, a search was carried out in the literature regarding the association of microRNAs in pre-neoplastic lesions in cervical cancer. The research was carried out in databases such as Pubmed and Science Direct, and scientific articles with publication date between the years 2010 to 2021 were selected and analyzed. , through bioinformatics tools. According to the inclusion and non-inclusion selection criteria, 8 articles were included in the literature review, which totaled 22 deregulated microRNAs in cervical intraepithelial lesions, with 18 microRNAs in high-grade lesions and 4 microRNAs in low-grade lesions. In silico analysis revealed that these microRNAs regulate genes such as: FZD7, ESR1 and BRAF, which are involved in major carcinogenesis pathways, such as the proteoglycan and stem cell pathway. The data presented demonstrate the deregulation of these biomolecules in pre-malignant cervical cancer samples.

Su desregulación se ha estudiado ampliamente en el cáncer, pero se ha documentado poco en lesiones premalignas, como las neoplasias intraepiteliales cervicales (NIC). En este contexto, este estudio tiene como objetivo buscar en la literatura microRNAs asociados a NICs, así como realizar predicciones in silico, con el fin de conocer los principales genes y vías que están regulados por ellos. Para ello se realizó una búsqueda en la literatura respecto a la asociación de microRNAs en lesiones preneoplásicas en cáncer de cérvix. La investigación se realizó en bases de datos como Pubmed y Science Direct, y se seleccionaron y analizaron artículos científicos con fecha de publicación entre los años 2010 al 2021, a través de herramientas bioinformáticas. De acuerdo con los criterios de selección de inclusión y no inclusión, se incluyeron 8 artículos en la revisión de la literatura, que totalizaron 22 microARN desregulados en lesiones intraepiteliales cervicales, con 18 microARN en lesiones de alto grado y 4 microARN en lesiones de bajo grado. El análisis in silico reveló que estos microARN regulan genes como: FZD7, ESR1 y BRAF, que están involucrados en las principales vías de carcinogénesis, como la vía de los proteoglicanos y las células madres. Los datos presentados demuestran la desregulación de estas biomoléculas en muestras premalignas de cáncer de cuello uterino.Os microRNAs são moléculas de RNA de fita simples, não-codificantes de proteínas, com cerca de 18-25 nucleotídeos, os quais são capazes de regular a expressão a nível pós-transcricional. A sua desregulação tem sido largamente estudada em câncer, porém pouco tem sido documentada em lesões pré-malignas, como as Neoplasias Intraepiteliais Cervicais (NIC). Neste contexto, este estudo tem como objetivo fazer uma busca na literatura dos microRNAs associados a NICs, assim como realizar a predição in silico, a fim de conhecer os principais genes e vias que são regulados por estes. Para isso, foi feita uma busca na literatura quanto à associação dos microRNAs em lesões pré-neoplásicas em câncer cervical. A pesquisa foi realizada em bases de dados, como o Pubmed e Science Direct, e foram selecionados e analisados artigos científicos com data de publicação entre os anos de 2010 a 2021. Em seguida, foi feita uma pesquisa dos genes alvos e vias envolvidas dos microRNAs, através de ferramentas de bioinformática. De acordo com os critérios de seleção de inclusão e não inclusão, foram incluídos na revisão de literatura 8 artigos, os quais totalizaram 22 microRNAs desregulados em lesões intraepiteliais cervicais, sendo 18 microRNAs em lesões de alto grau e 4 microRNAs de lesões de baixo grau. A análise in silico revelou que estes microRNAs regulam genes como: FZD7, ESR1 e BRAF, os quais estão envolvidos nas principais vias da carcinogênese, como a via dos proteoglicanos e células-tronco.
