Use of α and β-amyrin mixture as a storage medium in dental replantation

Resumo: The aim of the study was to evaluate the periodontal repair process of reimplanted rats teeth preserved in 10% α and β-amyrin solution. Maxillary right incisors of 36 rats were used, divided into 3 groups, the teeth were extracted and left in a dry medium and in the respective storage medium for 20 minutes and reimplanted. The groups were divided according to the conservation method used: SO - saline; AMSC - 10% α and β-amyrin solution, without endodontic treatment; AMCC - 10% solution of α and β-amyrin, with endodontic treatment. After 60 days, euthanasia was performed by transcardiac perfusion. The cuts obtained were stained in Hematoxylin and Eosin and Picrosirius Red. Extracellular matrix and blood vessels, nuclear profile, collagen fibers, organization of the periodontal ligament, extent and intensity of the inflammatory process, tooth resorption and ankylosis were evaluated. The Kruskal-Wallis test was used for comparison between groups. Among the significant differences, the apical third of the root in the AMCC group obtained a greater area of ​​blood vessels compared to SO, a smaller area of ​​collagen fibers of red birefringence and a greater area of ​​green fibers, in contrast, AMSC presented a greater area of ​​red birefringence fibers and smaller of green fibers. The other variables showed no difference between the groups. It is concluded that the mixture of α and β-amyrin in pre-clinical environment shown to have potential for use as a means of preservation in tooth avulsion.

Se utilizaron incisivos superiores derechos de 36 ratas, divididos en 3 grupos, se extrajeron los dientes y se dejaron en un medio seco y en el medio de conservación respectivo durante 20 minutos y reimplantaron. Los grupos se dividieron según el método de conservación utilizado: SO - salina; AMSC - solución de α y β-amirina 10%, sin tratamiento endodóntico; y AMCC - solución de α y β-amirina 10%, con tratamiento endodóntico. A los 60 días se realizó la eutanasia mediante perfusión transcardíaca. Los cortes obtenidos fueron teñidos con Hematoxilina y Eosina y Picrosirius Red. Se evaluaron matriz extracelular, vasos sanguíneos, perfil nuclear, fibras de colágeno, organización del ligamento periodontal, extensión e intensidad del proceso inflamatorio, reabsorción dentaria y anquilosis. Se utilizó la prueba de Kruskal-Wallis para la comparación. Entre las diferencias significativas, el tercio apical de la raíz en el grupo AMCC obtuvo mayor área de vasos sanguíneos en comparación con SO, menor área de fibras de colágeno de birrefringencia roja y mayor área de fibras verdes, en contraste, AMSC presentó mayor área de fibras de birrefringencia rojas y más pequeñas de fibras verdes. Las otras variables no mostraron diferencias entre los grupos. Se concluye que la mezcla de α y β-amirina en un entorno preclínico ha mostrado potencial para su uso como medio de conservación en la avulsión dental.O objetivo do estudo foi avaliar o processo de reparo periodontal de dentes de ratos reimplantados conservados em solução de α e β-amirina a 10%. A divisão dos grupos foi de acordo com o meio de conservação utilizado: SO – soro fisiológico 0,9%; AMSC - solução de α e β-amirina a 10%, sem tratamento endodôntico; e AMCC - solução de α e β-amirina a 10%, com tratamento endodôntico. Após 60 dias foi realizada eutanásia por perfusão transcardíaca. Os cortes obtidos foram corados em Hematoxilina e Eosina e Picrosirius Red. Foi avaliada matriz extracelular e vasos sanguíneos, perfil nuclear, fibras colágenas, organização do ligamento periodontal, extensão e intensidade do processo inflamatório, reabsorção dentária e anquilose. O teste de Kruskal-Wallis foi utilizado para comparação entre grupos. Dentre as diferenças significativas, o terço apical da raiz no grupo AMCC obteve maior área de vasos sanguíneos em comparação a SO, menor área de fibras colágenas de birrefringência vermelha e maior de fibras verdes, ao contrário, AMSC apresentou maior área de fibras de birrefringência vermelha e menor de fibras verdes. As outras variáveis não apresentaram diferença entre os grupos.
