Anti-inflamatory response of poultry challenged with LPS of Escherichia coli

Resumo: To evaluate the anti-inflammatory response of poultry, body weight (BW), body weight loss (BWL), cloacal temperature (CT) and expression of interleukin-10 (IL-10) after LPS application were calculated. A total of 19-day-old male broilers were used. At 17 days, the broilers were weighed and distributed in 12 metal cages with 1 animal per experimental unit. Intraperitoneal application of LPS was realized through 1 ml/kg of body weight at 19 days of age at time zero. For each treatment, that is, each time after the application of the LPS, the temperature of the poultry was measured. Afterwards, this chicken was weighed to determine body weight and body weight loss. Finally, the broiler was euthanized by cervical dislocation and collected their spleens for analysis of IL-10 mRNA expression. The analysis of the results obtained during the performance of this work concludes that the application of LPS of Escherichia coli directly interferes in the body weight loss of the animals (Y = - 0.09617 - 1.54780 x + 0.19107 x2) in 3.2307 % after 4.05 hours of application, and which directly interferes with the Gene Expression of the Interleukin-10 (Y = 1.33367 + 3.23013 x - 0.31733 x2), having its peak of expession after 5,0895 hours of application with a concentration of 9,53364 A.U..

Se utilizaron doce pollos de engorde de 19 días. A los 17 días, las aves fueron pesadas y distribuidas en 12 jaulas metálicas con 1 animal por unidad experimental. La aplicación intraperitoneal de LPS se realizó a una dosis de 1 ml / kg de peso vivo de aves a los 19 días de edad en el momento cero. Para cada tratamiento, es decir, cada vez después de aplicar el LPS, se midió la temperatura de las aves. Posteriormente, se pesó esta ave para determinar el peso final y la pérdida de peso. Finalmente, se sacrificó el ave y se recogió su bazo para el análisis de la expresión de ARNm de IL-10. El análisis de los resultados obtenidos durante la realización de este trabajo concluye que la aplicación de LPS de Escherichia coli interfiere directamente en la pérdida de peso de los animales (Y = - 0.09617 - 1.54780 x + 0.19107 x2) hasta 3.2307 % después de 4.05 horas de aplicación, y que interfiere directamente con la expresión génica de la citocina antiinflamatoria Interleucina-10 (Y = 1.33367 + 3.23013 x - 0.31733 x2), que tiene su pico de expresión después de 5, 0895 horas de aplicación con una concentración de 9,53361 A.U..Para avaliar a resposta anti-inflamatória de frangos de corte foram calculados peso final (PF), perda de peso (PP), temperatura retal (TR) e expressão de interleucina-10 (IL-10) após a aplicação de LPS. Foram utilizados 12 frangos de corte machos com 19 dias de idade. Aos 17 dias, as aves foram pesadas e distribuídas em 12 gaiolas de metal com 1 animal por unidade experimental. Foi realizada a aplicação intraperitoneal do LPS na dose de 1 ml/kg de peso vivo das aves aos 19 dias de idade no tempo zero. Para cada tratamento, ou seja, cada tempo após a aplicação do LPS, foi mensurada a temperatura das aves. Após, essa ave foi pesada para determinação de Peso Final e Perda de Peso. Finalmente, a ave foi abatida e realizada a coleta de seu baço para análise de expressão de RNAm de IL-10.
