Analytical methods for evaluating the stability of rutin and analyzing the formation of its degradation products: a review

Resumo: Rutin (RUT) is a flavonoid found in a wide variety of vegetables. It has multiple therapeutic properties, being especially indicated as a venotonic for the treatment of chronic venous insufficiency (CVI). To obtain medicines composed by RUT or associated with other drugs, it is necessary to initially evaluate the stability of the active pharmaceutical ingredient (API), with the objective guarantee the effectiveness and quality of the products, as well as the safety of consumers. Despite being a presupposition for drug registration, there is a scarcity of data in the literature regarding the stability of the API of RUT and its derived products, so this work aimed to perform a critical analysis of publications that address this issue. In view of the prospection, it was possible to divide the information into three groups: evaluation of the stability resulting from the extractive process in plant derivatives, of release systems developed with the purpose of circumventing intrinsic physicochemical limitations of the molecule and in reactive media and environmental conditions designed to degrade the molecule. The results showed that stability studies are challenging from a technical and analytical point of view, since for conducting forced degradation tests and developing stability indicative methods (SIMs), despite the existence of guides, resolutions, and international compendia with the guidelines for guiding the tests, each molecule has a unique chemical nature. Therefore, it is essential to build a stability profile, to propose the control of processes and the development of formulations that contain adjuvants that allow the formation of more stable products.

Tiene múltiples propiedades terapéuticas, estando especialmente indicado como venotónico para el tratamiento de la insuficiencia venosa crónica (IVC). Para la obtención de medicamentos compuestos por RUT o asociados a otros fármacos, es necesario evaluar inicialmente la estabilidad del principio activo farmacéutico (PAF), con el fin de garantizar la eficacia y calidad de los productos, así como la seguridad de los consumidores. A pesar de ser un presupuesto para el registro de medicamentos, existe escasez de datos en la literatura con respecto a la estabilidad de la PAF de RUT y sus productos derivados, por lo que este trabajo tuvo como objetivo realizar un análisis crítico de las publicaciones que abordan este tema. En vista de la prospección, fue posible dividir la información en tres grupos: evaluación de la estabilidad resultante del proceso extractivo en derivados vegetales, de sistemas de liberación desarrollados con el fin de sortear limitaciones fisicoquímicas intrínsecas de la molécula y en medios reactivos y condiciones ambientales diseñadas para degradar la molécula. Los resultados mostraron que los estudios de estabilidad son un reto desde el punto de vista técnico y analítico, ya que para la realización de pruebas de degradación forzada y el desarrollo de métodos indicativos de estabilidad (MIEs), a pesar de la existencia de guías, resoluciones y compendios internacionales con los lineamientos para orientar las pruebas, cada molécula tiene una naturaleza química única. Por lo tanto, es fundamental construir un perfil de estabilidad para proponer el control de procesos y el desarrollo de formulaciones que contengan adyuvantes que permitan la formación de productos más estables.A rutina (RUT) é um flavonóide encontrado em uma ampla variedade de vegetais. Possui múltiplas propriedades terapêuticas, sendo especialmente indicada como venotônico para o tratamento da insuficiência venosa crônica (IVC). Para obtenção de medicamentos compostos pela RUT ou associados com outros fármacos é necessário avaliar inicialmente a estabilidade do insumo farmacêutico ativo (IFA), a fim de garantir a eficácia e qualidade dos produtos, bem como a segurança dos consumidores. Apesar de ser um pressuposto para o registro de medicamentos, há na literatura escassez de dados referentes a estabilidade do IFA de RUT e seus produtos derivados, portanto este trabalho teve como objetivo realizar uma análise crítica das publicações que abordam essa temática. Diante da prospecção foi possível dividir as informações em três grupos: avaliação da estabilidade decorrida do processo extrativo em derivados vegetais, de sistemas de liberação desenvolvidos com a finalidade de contornar limitações físico-químicas intrínsecas da molécula e em meios reativos e condições ambientais projetados para degradar a molécula. Os resultados demonstraram que os estudos de estabilidade são desafiadores do ponto de vista técnico e analítico, uma vez que para condução dos testes de degradação forçada e desenvolvimento de métodos indicativos de estabilidade (MIEs), apesar da existência de guias, resoluções e compêndios internacionais com as diretrizes para norteamento dos ensaios, cada molécula tem uma natureza química única.
