In vitro antagonistic activity of Sclerotium sp. by Bacillus sp.

Resumo: Bacteria from the genus Bacillus sp. have a high potential for the biological control of phytopathogenic fungi. Therefore, in the present research we proposed to evaluate the antagonistic capacity of Bacillus sp. isolates against the phytopathogenic fungus Sclerotium sp. The tests were performed using two methods (dual and modified dual), also using five Bacillus sp. isolates (LBM01, LBM02, LBM03, LBM04 and LBM06). Mycelial growth evaluations were performed on BDA culture medium. All experiments were performed in triplicate and the plates were incubated under a 12 h photoperiod at 28±1 ºC for 7 days. Radial growth of the pathogens was measured at the intervals of 24, 48, 72, 96, 120, 144 and 168 h. The percentage of inhibition was calculated using the equation: I% = [(C-T)/C]x100. All Bacillus sp. isolates tested were able to inhibit the mycelial growth of Sclerotium sp. within 72 h, while isolates LBM01, LBM03 and LBM06 were able to maintain inhibition for 168 h. The isolate LBM06 showed the highest percentage of inhibition in all two culture methods used.

Por lo tanto, en la presente investigación nos propusimos evaluar la capacidad antagónica de los aislados de Bacillus sp. contra el hongo fitopatógeno Sclerotium sp. Las pruebas se realizaron utilizando dos métodos (dual y dual modificado), así como cinco aislados de Bacillus sp. (LBM01, LBM02, LBM03, LBM04 y LBM06). Las evaluaciones del crecimiento micelial se realizaron en medio de cultivo BDA. Todos los experimentos se realizaron por triplicado y las placas se incubaron bajo un fotoperiodo de 12 h a 28±1 ºC durante 7 días. El crecimiento radial de los patógenos se midió a intervalos de 24, 48, 72, 96, 120, 144 y 168 horas. El porcentaje de inhibición se calculó mediante la ecuación: I% = [(C-T)/C]x100. Todos los aislados de Bacillus sp. probados fueron capaces de inhibir el crecimiento micelial de Sclerotium sp. en 72 h, mientras que los aislados LBM01, LBM03 y LBM06 fueron capaces de mantener la inhibición durante 168 h. El aislado LBM06 mostró el mayor porcentaje de inhibición en todos los dos métodos de cultivo utilizados.As bactérias pertencentes ao gênero Bacillus sp. têm alto potencial para o controle biológico de fungos fitopatogênicos. Portanto, na presente pesquisa nos propusemos avaliar a capacidade antagônica de isolados de Bacillus sp. contra o fungo fitopatogênico Sclerotium sp. Os testes foram realizados utilizando dois métodos (dual e dual modificado), assim como cinco isolados de Bacillus sp. (LBM01, LBM02, LBM03, LBM04 e LBM06). As avaliações do crescimento micelial foram realizadas no meio de cultura BDA. Todos os experimentos foram realizados em triplicata e as placas foram incubadas sob fotoperíodo de 12 h a 28±1 ºC durante 7 dias. O crescimento radial dos patógenos foi medido nos intervalos de 24, 48, 72, 96, 120, 144 e 168 horas. A porcentagem de inibição foi calculada através da equação: I% = [(C-T)/C]x100. Todos os isolados de Bacillus sp. testados foram capazes de inibir o crescimento micelial do Sclerotium sp. em até 72 h, enquanto os isolados LBM01, LBM03 e LBM06 foram capazes de manter a inibição durante 168 h.
