Resumo: In the present study, we evaluated the effects of maternal exposure to glyphosate, during pregnancy and lactation period, in hepatic lipid metabolism on adult mice offspring with high-fat diet-fed. Thus, female mice C57BL/6 were randomly divided in: 1) Control group (CTL), which received standard chow and pure water ad libitum and 2) Glyphosate group (GF), which received standard chow and water with 0.5% of commercial formulation of GF (Roundup Original DI®). The treatment occurred from day 0 at 30th postnatal day, keeping the same food supplementation. At 60th postnatal day, offspring was separated in four groups: CTL group animals were subdivided in 1a) CTL-DP-F1 group, which kept same food supplementation and 1b) CTL-DH-F1 group, which received high-fat diet; GF group animals were subdivided in 2a) GF-DP-F1 group, which kept the same food supplementation and 2b) GF-DH-F1 group, which received a high-fat diet. At 150 days of life, offspring were euthanized. Body weight gain were similar between CTL-DP-F1 and GF-DP-F1, however lower than high-fat diet-fed groups. Nevertheless, this parameter was significantly lower in GF-DH-F1 groups when compared to CTL-DH-F1. High-fat diet consumption increased fat pad mass in CTL-DH-F1 group in relation to others groups at 150 days of age. GF-DH-F1 group increased 75 and 60% retroperitoneal and perigonadal fat pads, respectively, in relation to CTL-DP-F1 and GF-DP-F1. Interestingly, fat pad accumulation was 50% lower in GF-DH-F1 group when compared to CTL-DH-F1 group. CHOL and TG fasting serum levels were similar between CTL-DP-F1 and GF-DP-F1 and significantly lower to CTL DH-F1. Liver weight were higher in GF-DP-F1 group when compared to CTL DP-F1. Stands out that liver weight in GF-DH-F1 were higher in relation to CTL DH-F1. Liver total lipids content was similar between CTL-DP-FQ and GF-DP F1, but the TG content was higher in GF-DP-F1 group. GF-DH-F1 shows liver total lipids content and TG lower in relation to GF-DP-F1. The groups that received high-fat diet showed steatosis on liver. It was observed fibrosis on liver of 66.67% in CTL-DH-F1 and 40% in GF-DH-F1. All the treatment groups had lobular inflammation. Thus, it was show for the first time that maternal exposure to glyphosate in male mice leads to hepatic inflammation on adult offspring. When we analyzed the offspring which received high-fat diet, we observed that puppies which mothers were exposed to glyphosate had fat pad accumulation on white adipose tissue and liver, as well as soft dyslipidemia in relation to control dams offspring which received high-fat-diet.

Para isso, camundongos fêmeas adultos da linhagem C57BL/6 foram isoladas, após constatada a prenhez, e separadas em dois grandes grupos: 1) Grupo Controle (CTL), que recebeu dieta padrão e água ad libitum e 2) Grupo Glifosato (GF), que recebeu dieta padrão e água com 0,5% da formulação comercial de glifosato (Roundup Original DI®). O tratamento ocorreu do dia 0 até o 30º dia pós-natal, mantendo a mesma suplementação alimentar. No 60° dia pós-natal, a prole foi separada em quatro grupos: os animais do grupo CTL foram subdivididos em 1a) Grupo CTL-DP-F1, que permaneceram com a mesma suplementação alimentar e 1b) Grupo CTL-DH-F1, que recebeu dieta com alto teor de gordura; os animais do grupo GF foram subdivididos em 2a) Grupo GF-DP-F1, que continuaram recebendo a dieta padrão e 2b) Grupo GF-DH-F1, que receberam dieta com alto teor de gordura. Aos 150 dias de vida os animais sofreram eutanásia e foram coletadas amostras biológicas para análise plasmática e histológica. O ganho de peso corporal foi similar entre os animais do grupo CTL-DP-F1 e GF DP-F1, porém menor em relação aos animais que ingeriram dieta hiperlipídica. Todavia este parâmetro foi significativamente menor nos animais do grupo GF DH-F1 em relação aos CTL-DH-F1. O acúmulo de gordura aumentou nos animais do grupo CTL-DH-F1 em relação aos demais grupos aos 150 dias. Nos animais do grupo GF-DH-F1, a dieta aumentou em 75 e 60% o peso das gorduras retroperitoneal e perigonadal, respectivamente, em relação aos grupos CTL-DP-F1 e GF-DP-F1. Interessantemente, o acúmulo de gordura foi aproximadamente 50% menor nos animais GF-DH-F1 em relação ao grupo CTL-DH-F1. As concentrações plasmáticas de COL e TG foram similares entre os grupos CTL-DP-F1 e GF-DP-F1 e significativamente menores em relação ao grupo CTL-DH-F1. O peso do fígado foi maior no grupo GF-DP-F1 quando comparado ao CTL-DP-F1. Destaca-se que o peso do fígado dos animais GF DH-F1 foi maior em relação ao grupo CTL-DH-F1. O conteúdo total de lipídios do fígado foi semelhante entre os grupos CTL-DP-F1 e GF-DP-F1, diferentemente do conteúdo de triglicerídeos que foi significativamente maior nos animais do grupo GF-DP-F1. O grupo GF-DH-F1 apresentou conteúdo total de lipídios e de TG no fígado, menores em relação ao grupo GF-DP-F1. Os fígados dos animais que receberam a dieta hiperlipídica apresentaram esteatose. Foi observada fibrose em 66,67% dos animais do grupo CTL-DH-F1 e em 40% do grupo GF-DH-F1. Enquanto nos animais do grupo CTL-DP-F1 não foram observados focos inflamatórios no tecido hepático, nos demais grupos foram observados infiltrados de leucócitos. Assim, demonstramos pela primeira vez que a exposição materna de camundongos machos ao glifosato leva à inflamação hepática na prole adulta. description In the present study, we evaluated the effects of maternal exposure to glyphosate, during pregnancy and lactation period, in hepatic lipid metabolism on adult mice offspring with high-fat diet-fed. Thus, female mice C57BL/6 were randomly divided in: 1) Control group (CTL), which received standard chow and pure water ad libitum and 2) Glyphosate group (GF), which received standard chow and water with 0.5% of commercial formulation of GF (Roundup Original DI®). The treatment occurred from day 0 at 30th postnatal day, keeping the same food supplementation. At 60th postnatal day, offspring was separated in four groups: CTL group animals were subdivided in 1a) CTL-DP-F1 group, which kept same food supplementation and 1b) CTL-DH-F1 group, which received high-fat diet; GF group animals were subdivided in 2a) GF-DP-F1 group, which kept the same food supplementation and 2b) GF-DH-F1 group, which received a high-fat diet. At 150 days of life, offspring were euthanized. Body weight gain were similar between CTL-DP-F1 and GF-DP-F1, however lower than high-fat diet-fed groups. Nevertheless, this parameter was significantly lower in GF-DH-F1 groups when compared to CTL-DH-F1. High-fat diet consumption increased fat pad mass in CTL-DH-F1 group in relation to others groups at 150 days of age. GF-DH-F1 group increased 75 and 60% retroperitoneal and perigonadal fat pads, respectively, in relation to CTL-DP-F1 and GF-DP-F1. Interestingly, fat pad accumulation was 50% lower in GF-DH-F1 group when compared to CTL-DH-F1 group. CHOL and TG fasting serum levels were similar between CTL-DP-F1 and GF-DP-F1 and significantly lower to CTL DH-F1. Liver weight were higher in GF-DP-F1 group when compared to CTL DP-F1. Stands out that liver weight in GF-DH-F1 were higher in relation to CTL DH-F1. Liver total lipids content was similar between CTL-DP-FQ and GF-DP F1, but the TG content was higher in GF-DP-F1 group. GF-DH-F1 shows liver total lipids content and TG lower in relation to GF-DP-F1. The groups that received high-fat diet showed steatosis on liver. It was observed fibrosis on liver of 66.67% in CTL-DH-F1 and 40% in GF-DH-F1. All the treatment groups had lobular inflammation. Thus, it was show for the first time that maternal exposure to glyphosate in male mice leads to hepatic inflammation on adult offspring. When we analyzed the offspring which received high-fat diet, we observed that puppies which mothers were exposed to glyphosate had fat pad accumulation on white adipose tissue and liver, as well as soft dyslipidemia in relation to control dams offspring which received high-fat-diet.
