Resumo: The objective of this work was to plan the synthesis of ZIF-8 and carry out its characterization, as well as carry out the characterization of vitexin (VIT) and to obtain and characterize (physical-chemical and pharmacological) a system based on vitexin and ZIF-8. For the synthesis of ZIF-8, a 32 factorial design was used and the ZIF-8 was characterized by techniques such as FT-IR, TG, DSC and SEM. VIT was obtained from the leaves of Jatropha mutabilis and was characterized by the techniques already mentioned; UV-vis and HPLC-DAD calibration curves were obtained, their release profile was evaluated, and their quantitative solubility was verified. For the synthesis of VIT@ZIF-8 system, a 22 factorial design was used; the characterization was performed by the tests mentioned above and the pharmacological action was verified in a murine model of primary dysmenorrhea. Among the reaction conditions used for the synthesis of ZIF-8, the one that provided the best yield was the one with reagents in a 1:2 ratio and occurred for 12 h; FT-IR analysis showed the characteristic bands present in 2-methylimidazole; SEM analyzes revealed the standard appearance of the MOF under study; TG analysis revealed mass loss thermal events related to water desorption, degradation of unbound 2-methylimidazole molecules and degradation of ZIF-8; the ZIF-8 DSC curve revealed no signs. Regarding vitexin, its structure was confirmed by NMR and FT-IR, with the presence of published characteristic signals and bands; its solubility proved to be low; the constructed calibration curves were linear, and their release did not exceed 35%; TG analysis revealed mass loss thermal events related to adsorbed water loss, melting and degradation; the DSC curve revealed a single endothermic event related to its fusion; SEM analysis revealed crystal morphology into spheres of different sizes. On VIT@ZIF-8, it was evidenced that the conditions that provide the best results for yield and incorporation rate were achieved at high levels; FT-IR revealed characteristic signals for VIT and ZIF-8, in addition to a new signal at 1743 cm-1 that can be considered evidence of system formation; TG and DSC data evidenced the characteristic events for both test substances, and a new thermal pattern was revealed for VIT@ZIF-8; the release test revealed that VIT@ZIF-8 releases 100% of the vitexin incorporated into the net within 1 min; regarding the pharmacological action, vitexin and VIT@ZIF-8 were able to reduce abdominal contortions orally at concentrations of 3 and 30 mg/kg, however, intraperitoneally this effect was only observed at a concentration of 30 mg/ kg. The work brought new data, which reaffirms its importance and originality.

Para síntese da ZIF-8, foi empregado um planejamento fatorial 32 e a ZIF-8 foi caracterizada por técnicas como FT-IR, TG, DSC e MEV. VIT foi obtida das folhas de Jatropha mutabilis e foi caracterizada pelas técnicas já mencionadas; curvas de calibração por UV-vis e CLAE-DAD foram obtidas, seu perfil de liberação foi avaliado, bem como sua solubilidade quantitativa foi verificada. Para a síntese do sistema VIT@ZIF-8, um planejamento fatorial 22 foi empregado; a caracterização foi realizada pelos testes já mencionados e a ação farmacológica foi verificada em modelo murino de dismenorreia primária. Dentre as condições de reações empregadas para a síntese da ZIF-8, a que forneceu melhor rendimento foi a que possui reagentes na proporção 1:2 e ocorreu durante 12 h; as análises por FT-IR mostraram as bandas características presentes no 2-metilimidazol; análises por MEV revelaram o aspecto padrão da MOF em estudo; a análise por TG revelou eventos térmicos de perda de massa referentes a dessorção da água, degradação de moléculas do 2-metilimidazol não ligadas e degradação da ZIF-8; a curva DSC da ZIF-8 não revelou sinais. No que tange a vitexina, sua estrutura foi confirmada por RMN e FT-IR, com presença de sinais e bandas características já publicadas; sua solubilidade revelou-se baixa; as curvas de calibração construídas mostraram-se lineares, e sua liberação não excedeu 35%; a análise por TG revelou eventos térmicos de perda de massa relacionados a perda de água adsorvida, fusão e degradação; a curva DSC revelou um único evento endotérmico relacionado a sua fusão; a análise por MEV revelou morfologia de cristais em formatos de esferas de diferentes tamanhos. Sobre VIT@ZIF-8, ficou evidenciado que as condições que fornecem os melhores resultados para rendimento e taxa de incorporação foram conseguidas em níveis altos; o FT-IR revelou sinais característicos para VIT e para ZIF-8, além de um novo sinal em 1743 cm-1 que pode ser considerada uma evidência da formação do sistema; os dados do TG e DSC evidenciaram os eventos característicos para ambas as substâncias-teste, e um novo padrão térmico foi revelado para VIT@ZIF-8; o teste de liberação revelou que VIT@ZIF-8 libera 100% da vitexina incorporada na rede em 1 min; a respeito da ação farmacológica, vitexina e VIT@ZIF-8 foram capazes de reduzir as contorções abdominais por via oral nas concentrações de 3 e 30 mg/kg, no entanto, por via intraperitoneal esse efeito só foi observado na concentração de 30 mg/kg.
