Detecção e caracterização molecular de Sarcocystis spp. em tecidos de ovinos abatidas em frigoríficos no Sul do Brasil

Resumo: Sarcocystis spp. are intracellular protozoan parasites with an intermediate-definitive host life cycle and can infect a wide range of animals. There are some differences among the Sarcocystis species related to the life cycle and pathogenicity, but the formation of tissue cysts is a common characteristic within the genus. In sheep, Sarcocystis species that commonly cause infection are Sarcocystis tenella, Sarcocystis arieticanis, Sarcocystis gigantea and Sarcocystis medusiformis. S tenella and S. arieticanis present canids as definitive hosts, form microscopic cysts and can cause clinical disease or subclinical infection. On the other hand, S. gigantea and S. medusiformis which have felids as definitive hosts form macroscopic cysts and, although these Sarocystis species do not have pathogenic potential in sheep, the presence of macrocysts cause condemnation of sheep meat and organs. Sarcocystosis is a widespread infection, with high occurrence rates mainly in developing countries. Although some studies have already demonstrated the presence of Sarcocystis species in sheep breeding in Brazil, more knowledge about sheep Sarcocystosis epidemiology is essential. Therefore, the aims of this study were to investigate the occurrence of Sarcocystis spp. in sheep slaughtered in Santa Maria and Santiago, as well as to check the presence of Sarcocystis spp. DNA in the evaluated samples and to identificate the Sarcocystis species through genetic sequencing. A part of the18S rDNA gene was selected for PCR amplification, followed by genetic sequencing aiming Sarcocystis species identification. Ten of the 130 sheep had macrocysts and all of these macrocysts were confirmed as S. gigantea. Micro cysts were found in 125 animals. All microcysts were of the Sarcocystis genus as shown by PCR amplification. Ten of these samples, randomly selected for gene sequencing, were confirmed as S. tenella. The frequency of microscopic cysts was high, although this is not surprising. Macroscopic cysts were found only in the esophagus. This study presents the first molecular confirmation of the presence of S. gigantea in Brazil.

Dentre as espécies de Sarcocystis, existem algumas diferenças em relação ao ciclo e patogenicidade, porém a formação de cistos teciduais é uma característica comum dentro do gênero. Em ovinos, as espécies de Sarcocystis que comumente estabelecem infecção são: Sarcocystis tenella e Sarcocystis arieticanis que apresentam a característica de formar cistos microscópicos e podem causar doença clínica ou infecção subclínica nesses animais, e Sarcocystis gigantea e Sarcocystis medusiformis formam cistos macroscópicos e apesar de não apresentarem potencial patogênico, podem acarretar relevantes perdas econômicas relacionadas a condenações de órgãos e carcaças em frigoríficos. As duas primeiras espécies apresentam canídeos como hospedeiros definitivos, enquanto as duas últimas tem como hospedeiros definitivos os felídeos. A sarcocistose é uma infecção disseminada por todo o mundo, apresentando altas taxas de ocorrência principalmente em países em desenvolvimento. No Brasil apesar de alguns estudos já demostrarem a presenças de Sarcocystis nos ovinos, os dados ainda são escassos e a epidemiologia da doença ainda é pouco conhecida. Tendo em vista a importância da infecção por Sarcocystis spp. nos ovinos, e a fim de preencher as lacunas existentes sobre essa questão este trabalho foi desenvolvido com intuito de pesquisar a ocorrência de Sarcocystis spp. em ovinos abatidos em Santa Maria e em Santiago, bem como, verificar a presença de DNA de Sarcocystis spp. nas amostras avaliadas, e identificar as espécies de Sarcocystis spp. nas amostras através da técnica de sequenciamento genético. Para a realização da reação de PCR foi selecionado o gene 18S rDNA, seguido de sequenciamento de amostras para identificar as diferentes espécies de Sarcocystis spp. Das 130 amostras analisadas, 10 foram positivas para presença de cistos macroscópicos, onde através da PCR se constatou S. gigantea, e este resultado foi confirmado pelo sequenciamento genético. Enquanto, cistos microscópicos foram encontrados em 125 animais, onde a PCR identificou a presença de S. tenella. Destas 125 amostras, 10 foram submetidas ao sequenciamento genético que confirmou a presença de S. tenella. A frequência de cistos microscópicos foi alta, embora esse não seja um dado surpreendente. Os cistos macroscópicos foram encontrados apenas no esôfago.
