Diversidade genética estimada por meio de marcadores moleculares e morfológicos em acessos de mini-tomate

Resumo: The tomato is the second most cultivated and consumed vegetables in the world and is among the most genetically studied plant species and phenotypically modified. Tomato production has grown in recent years, and this growth is due to the introduction of new cultivars and varieties such as mini-tomatoes, developed by breeding programs. In prebreeding programs the characterization of accessions of a germoplasm bank is essential to determine the usefulness of the accessions as a source of genes of interest. This paper describes the establishment of mini-tomato UNICENTRO germplasm bank and the characterization of this germplasm bank by means of molecular markers, morphological characters, agronomic performance and fruit color. For molecular analysis was used 11 ISSR (Inter-simple sequence repeat) primers. The polymorphism of ISSR was 88%, considered high for tomato species. Based on the results of the PIC (polymorphism information content), MI (markers index) and RP (resolving power) primers 878, 861, 807, 815 and 836 are the most informative and the most efficient in the discrimination of accessions, therefore the most suitable for studies with mini-tomatoes.. The average similarity between accessions was 64%, accesses RVTC-15 and IAC420-62 were the least similar (13%) and accessions RVTC-25 and RVTC-26 the most similar (96%). The 6 and 8 groups generated in the dendogram (molecular and morphological) showed no relationship with the local of sampling. With morphological data was obtained average standardized distance between access of the 0.60. The access IAC1498-63 was show the higher distance between any access to morphological characteristics. Although it is difficult to make a direct comparison between similarity / divergence, obtained with molecular markers with genetic distance, standardized distances (in values ranging from 0 to 1) made in this work, allows a more direct comparison and therefore conclude that iii the morphological markers, compared to molecular markers, demonstrated greater capacity to differentiate genotypes.. As for agronomic performance, access RVTC-22 is the genotype that most stands out for good fruit production and high soluble solids. However, considering the different criteria for fruit color, accesses IAC1622-64 and IAC392-59 were also important and may indicate these three accessions for breeding programs of mini-tomatoes. Based on these results it can be concluded that the accessions showed good values of divergence molecular and morphoagronomic to all features, so these accessions are potentially useful in breeding programs of minitomatoes.

A produção de tomate tem crescido nos últimos anos devido à introdução de novas cultivares, como os mini-tomates, desenvolvidas por programas de melhoramento. Em programas de pré-melhoramento a caracterização de acessos de um banco de germoplasma é essencial para determinar a utilidade dos acessos como fonte de genes de interesse. Neste trabalho é descrito a montagem do banco de germoplasma de minitomates da UNICENTRO bem como a caracterização deste banco por meio de marcadores moleculares, morfológicos, desempenho agronômico e coloração dos frutos. Para análise molecular foi utilizado 11 marcadores ISSR (Inter-Simple Sequence Repeat). O polimorfismo dos marcadores ISSR foi de 88%, considerado alto para espécies de tomateiro. Com base nos resultados do PIC (Conteúdo de Informação Polimórfica), MI (Ándice do Marcador) e RP (Poder de Resolução) os primers 878, 861, 807, 815 e 836 são os mais informativos e os mais eficientes na discriminação dos acessos, sendo assim os mais indicados para estudos com mini-tomates. A similaridade média entre os acessos foi de 64%, os acessos RVTC-15 e IAC420-62 foram os menos similares (13%) e os acessos RVTC-25 e RVTC-26 os mais similares (96%). Os 6 e 8 grupos gerados no dendograma (molecular e morfológico) não apresentaram relação direta com os locais de coleta. Com os dados morfológicos foi obtido uma distância média padronizada entre os acessos de 0,60. O acesso IAC1498-63 foi o o que apresentou a maior distância entre todos os acessos para as características morfológicas. Embora seja difícil fazer uma comparação direta entre similaridade/divergência, obtida com os marcadores moleculares, com distância genética, a padronização das distâncias (em valores variando de 0 a 1) feita neste trabalho, permite uma comparação mais direta e consequentemente, concluir que os marcadores morfológicos, quando comparados com os marcadores moleculares, demonstraram maior capacidade de diferenciar os genótipos estudados. Quanto ao desempenho agronômico, o acesso RVTC-22 foi o genótipo que mais se destaca pela boa produção de frutos e elevado teor de sólidos solúveis. Contudo, considerando os diferentes critérios para coloração dos frutos, os acessos IAC1622-64 e IAC392-59 também se destacaram, podendo indicar esses três acessos para programas de melhoramento de mini-tomates. Com base nesses resultados ii pode-se concluir que os acessos apresentaram bons valores de divergência genética molecular e morfoagronômica para todas as características, assim, esses acessos possuem potencial para utilização em programas de melhoramento de mini-tomate.Made available in DSpace on 2016-09-20T12:12:32Z (GMT).
