Protocol for obtaining Cedrela odorata L. plants through tissue culture

Resumo: The work aimed to develop a basic protocol for micropropagation of Cedrela odorata L. plants from seedlings germinated in vitro. The experiments were conducted at the Embrapa Amazônia Oriental Genetic Resources and Biotechnology laboratory. For the disinfestation of seeds, sodium hypochlorite solution was tested at 0; 1.0; 1.5 and 2.0% for 10, 15 and 20 minutes. For budding induction in MS medium, 5 concentrations of BAP and 2 types of segments were tested and rooting was used 0; 1.0; 1.5; 2.0 and 2.5 mg.L-1 of ANA. The design used was completely randomized or in a factorial arrangement according to the treatments analyzed. The data obtained in the different experiments were subjected to analysis of variance and the means compared by the Tukey test at 5% probability. The results showed that the use of 2.0% sodium hypochlorite for 15 minutes showed the lowest percentage of contamination of the seeds. Treatment with 1 mg. L-1 of BAP promoted the largest number and length of shoots in apical segments and with the addition of 2 g.L-1 of sucrose induced the appearance of shoots in stem segments. The largest number and length of root / explant was obtained with 1 mg. ANA L-1. It is possible to obtain a complete Cedrella odorata plant from disinfected seeds in 2.0% sodium hypochlorite solution, and the use of 1 mg. L-1 of BAP and ANA for budding induction and rooting, respectively.

Los experimentos se realizaron en el laboratorio de Biotecnología y Recursos Genéticos de Embrapa Amazônia Oriental. Para la desinfestación de semillas, se probó la solución de hipoclorito de sodio a 0; 1,0; 1,5 y 2,0% durante 10, 15 y 20 minutos. Para la inducción de gemación en medio MS, se probaron 5 concentraciones de BAP y 2 tipos de segmentos y se utilizó enraizamiento 0; 1,0; 1,5; 2,0 y 2,5 mg. L-1 de ANA. El diseño utilizado fue completamente al azar o en arreglo factorial según los tratamientos analizados. Los datos obtenidos en los diferentes experimentos se sometieron a análisis de varianza y se compararon las medias mediante la prueba de Tukey al 5% de probabilidad. Los resultados mostraron que el uso de hipoclorito de sodio al 2.0% durante 15 minutos mostró el menor porcentaje de contaminación de las semillas. Tratamiento con 1 mg. L-1 de BAP promovió la mayor cantidad y longitud de brotes en segmentos apicales y con la adición de 2 g. L-1 de sacarosa indujo la aparición de brotes en segmentos de tallo. El mayor número y longitud de raíz / explante se obtuvo con 1 mg. ANA L-1. Es posible obtener una planta completa de Cedrella odorata a partir de semillas desinfectadas en solución de hipoclorito de sodio al 2,0% y el uso de 1 mg. L-1 de BAP y ANA para inducción de gemación y enraizamiento, respectivamente.     O trabalho teve como objetivo desenvolver um protocolo básico para micropropagação de plantas de Cedrela odorata L. a partir de plântulas germinadas in vitro. Os experimentos foram conduzidos no laboratório de Recursos Genéticos e Biotecnologia da Embrapa Amazônia Oriental. Para a desinfestação das sementes foram testadas solução de hipoclorito de sódio a 0; 1,0; 1,5 e 2,0% por 10, 15 e 20 minutos. Para indução de brotações em meio MS foi testada 5 concentrações de BAP e 2 tipos de segmentos e no enraizamento foi utilizado 0; 1,0; 1,5; 2,0 e 2,5 mg.L-1 de ANA. O delineamento utilizado foi inteiramente casualizado ou em arranjo fatorial de acordo com os tratamentos analisados. Os dados obtidos, nos diferentes experimentos foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey a 5% de probabilidade. Os resultados demonstraram que o uso de hipoclorito de sódio a 2,0% por 15 minutos apresentou a menor percentagem de contaminação das sementes. O tratamento com 1 mg. L-1 de BAP promoveu o maior número e comprimento de brotos em segmentos apicais e com acréscimo de 2 g.L-1 de sacarose induziu o surgimento de brotos em segmentos caulinares. O maior número e comprimento de raiz/explante foi obtido com 1 mg. L-1 de ANA. É possível obter planta completa de Cedrella odorata a partir de sementes desinfestadas em solução de hipoclorito de sódio a 2,0 %, e o uso de 1 mg.
