Analysis of the cytotoxic, genotoxic and mutagenic potential of the hydroalcoholic extract of Morus nigra L. Leaves through the bioassay Allium cepa

Summary: Consumption of products and substances of plant origin is common practice in populations all over the planet. Morus nigra L. (blackberry) is a medicinal plant widely used in folk medicine, and has several active ingredients with therapeutic action, such as antioxidant, antinoceptive, hypoglycemic and anti-inflammatory activity. The aim of the present study was to analyze the possible cytotoxic, genotoxic and mutagenic activities of Morus nigra L. leaves through the Allium cepa test. For preparation of the extract were used blackberry leaves, collected in the city of Agricolândia-PI. Three different concentrations (0.007 mg / mL, 0.07 mg / mL and 0.7 mg / mL) were used, in addition to the negative and positive controls. For each extract concentration, 10 onion bulbs were used, which were exposed for germination. After growth, the roots were measured with the help of a ruler, cut and followed by the slide preparation procedure, which were visualized under a light microscope at 100x magnification and analyzed qualitatively for the presence of genetic damage. Statistical analysis was performed using the ANOVA system of mean sample results. After data analysis, it was observed that the size of root meristems indicated no toxicity, the mitotic indexes of A. cepa cells exposed at these concentrations were not statistically different from the negative control. The chromosomal aberrations found were not significant (P <0.05) in the concentrations used and micronuclei and binucleated cells did not show significant variations when compared to the negative control results. Thus, based on the results, the use of Morus nigra leaves is safe at the concentrations studied, but some studies can be performed to confirm the results and suggest greater reliability in the use of this plant by the population safely.

Morus nigra L. (mora) es una planta medicinal ampliamente utilizada en medicina popular, y tiene varios ingredientes activos con acción terapéutica, como actividad antioxidante, antinoceptiva, hipoglucémica y antiinflamatoria. El objetivo del presente estudio fue analizar las posibles actividades citotóxicas, genotóxicas y mutagénicas de las hojas de Morus nigra L. a través de la prueba Allium cepa. Para la preparación del extracto se utilizaron hojas de mora, recolectadas en la ciudad de Agricolândia-PI. Se utilizaron tres concentraciones diferentes (0,007 mg / ml, 0,07 mg / ml y 0,7 mg / ml), además de los controles negativos y positivos. Para cada concentración de extracto, se utilizaron 10 bulbos de cebolla, que fueron expuestos para la germinación. Después del crecimiento, las raíces se midieron con la ayuda de una regla, se cortaron y luego se realizó el procedimiento de preparación del portaobjetos, que se visualizó bajo un microscopio óptico con un aumento de 100x y se analizó cualitativamente la presencia de daño genético. El análisis estadístico se realizó utilizando el sistema ANOVA de resultados de muestras medias. Después del análisis de los datos, se observó que el tamaño de los meristemos de la raíz no indicaba toxicidad, los índices mitóticos de las células de A. cepa expuestas a estas concentraciones no eran estadísticamente diferentes del control negativo. Las aberraciones cromosómicas encontradas no fueron significativas (P <0.05) en las concentraciones utilizadas y los micronúcleos y las células binucleadas no mostraron variaciones significativas en comparación con los resultados del control negativo. Por lo tanto, según los resultados, el uso de hojas de Morus nigra es seguro a las concentraciones estudiadas, pero se pueden realizar algunos estudios para confirmar los resultados y sugerir una mayor confiabilidad en el uso de esta planta por parte de la población de manera segura.O consumo de produtos e substâncias de origem vegetal é prática comum em ameaças de todo o planeta. O Objective do Presente Estudo foi analisar como Possíveis Atividades citotóxicas, genotóxicas e mutagênicas das Folhas da Morus nigra L. atraves do teste Allium cepa. Para preparar o extrato foram utilizados folhas de amora, coletadas na cidade de Agricolândia-PI. Foram usados três tipos diferentes (0,007 mg / mL, 0,07 mg / mL e 0,7 mg / mL), além do controle negativo e positivo. Para cada concentração de extrato, são utilizados 10 bulbos de cebola, que ficam expostos para germinação. Após o crescimento como raízes foram medidas com auxílio de régua, cortadas e seguidas, ou procedimento de preparação de lâminas, que foram visualizadas no microscópio óptico em um aumento de 100x e analisadas qualificativamente relativamente à presença de danos genéticos. Uma análise estatística foi feita através do sistema ANOVA da média dos resultados das amostras.
