Avaliação histológica de neoplasias mamárias induzidas por DMBA em camundongos SWISS sujeitos à dieta hiperlipídica e tratados com inibidor de COX-2

Resumo: The present study has as objective to report through a scientific article the comparison of two references to classify DMBA-induced breast tumors in female Swiss mice, as well as to evaluate and correlate the tissue inflammation in these tumors of mice subjected to a high fat diet (DH) and treatment with etoricoxib. In this work, breast cancer and animal models were reviewed, as well as the role of the lipid-rich diet and tissue inflammation in the neoplastic context. 52 tumors induced by DMBA in mice were histologically classified. Subsequently, only those of mammary origin were analyzed for inflammatory infiltrate and angiogenesis. 22/52 (42.31%) tumors of mammary origin were observed. From these, it was possible to classify 2 histological types according to Sass and Dunn, whereas according to Cassali, 7 types were obtained, and Adenoacantoma and Micropapillary Carcinoma were the most frequent type, respectively. Among the carcinomas, there were 3 more aggressive types, present significantly more frequently in the DH groups, and there was no difference in the group treated with etoricoxib. Likewise, there was a slight increase in the intensity of inflammatory infiltrate and number of blood vessels in the DH groups. Therefore, it is concluded that DMBA did not induce mammary tumors in the majority, and the histological classification of Cassali was more detailed. Although treatment with Etoricoxib did not decrease inflammation, the DH was able to induce a greater degree of tumor malignancy.

Although treatment with Etoricoxib did not decrease inflammation, the DH was able to induce a greater degree of tumor malignancy.O presente estudo tem por objetivo relatar através de artigo científico a comparação de duas referências para classificar tumores de mama induzidos por DMBA em camundongos Swiss fêmeas, bem como avaliar e correlacionar a inflamação tecidual nestes tumores de camundongos submetidas à dieta hiperlipídica (DH) e tratadas com etoricoxibe. Neste trabalho, revisaram-se o câncer de mama e os modelos animais, além do papel da dieta rica em lipídios e inflamação tecidual no contexto neoplásico. 52 tumores induzidos por DMBA em camundongos foram classificados histologicamente. Posteriormente, somente os de origem mamária foram analisados quanto ao infiltrado inflamatório e angiogênese. 22/52 (42,31%) tumores de origem mamária foram observados. Destes, foi possível classificar 2 tipos histológicos de acordo com Sass e Dunn, enquanto de acordo com Cassali obteve-se 7 tipos, destacando-se Adenoacantoma e Carcinoma Micropapilar como os mais frequentes, respectivamente. Dentre os carcinomas, observaram-se 3 tipos mais agressivos, presentes significativamente em maior frequência nos grupos com DH, não havendo diferença no grupo tratado com etoricoxibe. Da mesma forma, houve leve aumento da intensidade de infiltrado inflamatório e número de vasos sanguíneos nos grupos DH. Por isso, conclui-se que o DMBA não induziu majoritariamente tumores mamários e a classificação histológica de Cassali se mostrou mais detalhada.
