Canal de potencial receptor transiente vanilóide na termorregulação de mamíferos

Resumo: | The transient receptor potential channels (TRPs) became important in lipid metabolism and thermoregulation. TRPV1 has been related to the control of white adipocyte and brown adipose tissue (BAT) physiology, which has a high thermogenic activity, mainly through the uncoupling protein UCP1. Factors such as core body temperature, or feeding, are major modulating agents of local clocks in the metabolism centers as liver and adipose tissue. In the literature, a putative interaction of natriuretic peptides (NPs) with the regulation of the metabolic activity of the adipose tissue. In mice subject to cold, NPs affect the adipose tissue favoring thermogenesis in BAT. We proposed to evaluate the participation of the TRPV1 channel in the regulation of BAT thermogenic responses in C57Bl/6J (WT) e Trpv1 KO mice subject to thermoneutral and cold conditions. We determined the thermoneutral zone of both genotypes, which allowed us the choice of 30°C+1 as the control temperature. The lack of TRPV1 channel decreases the locomotor activity without affecting the core body temperature. With thermographic images, a more intense dissipation of heat by the body periphery was noted in WT animals in the thermoneutral condition. Upon chronic cold challenge of 22°C, Trpv1 KO mice exhibited higher BAT thermogenic activity and maintained their body temperature similar to the WT group. |

O TRPV1 está relacionado com controle da fisiologia dos adipócitos brancos, e do tecido adiposo marrom (BAT), que possui alta capacidade termogênica, através principalmente da proteína desacopladora UCP1. Fatores como a temperatura corpórea interna, ou alimentação, são importantes moduladores de relógios locais em centros de metabolismo e termogênese como fígado e tecido adiposo. Na literatura, foi demonstrada uma possível interação dos peptídeos natriuréticos (NPs) na regulação da atividade metabólica do tecido adiposo. Em camundongos submetidos ao frio, os NPs atuam no tecido adiposo favorecendo a termogênese no BAT. Decidimos então, avaliar a participação do canal TRPV1 na regulação das respostas termogênicas no BAT. Para tanto, utilizamos camundongos C57Bl/6J (WT) e Trpv1 KO em condição termoneutra e de frio crônico. Determinamos a zona de termoneutralidade de ambos, o que nos possibilitou a utilização de 30°C+1 como temperatura controle. A ausência do canal TRPV1 parece causar diminuição da atividade locomotora sem influenciar a temperatura corpórea interna. Através de imagens termográficas, notamos que animais WT dissipam maior quantidade de calor pela periferia do corpo em condição termoneutra.
