Avaliação radiográfica da periodontite experimental induzida pela porphyromonas gingivalis com e sem ligaduras após tratamento com alendronato e resveratrol

Resumo: Periodontitis is a chronic inflammatory disease that develops over time through the accumulation of bacterial biofilm, inducing the exacerbated production of immuneinflammatory mediators. The aim of this study was to radiographically analyze the influence of alendronate and resveratrol on periodontitis induced by Porphyromonas gingivalis (P. gingivalis) with and without ligation. One hundred and twenty rats were submitted to experimental periodontitis through two techniques of induction of the disease: with ligation associated with P. gingivalis (LPg) and without ligation associated with P. gingivalis (SLPg). After the disease was established, the rats were divided into 4 groups according to the treatment: the alendronate group (ALN), which received the dose of 2 mg/kg/day of alendronate; the resveratrol group (RESV) receiving the 10 mg/kg/day dose of resveratrol dissolved in 10 mg/ml soluble starch; β- cyclodextrin (β-CD) /RESV group receiving the 10 mg/kg/day dose of the complex and control group receiving the 10 mg/kg/day β- CD dose. The medications were administered orally by gavage for 30 days. The animals were euthanized and the jaw surgical specimens were x-rayed and submitted to ImageJ ® software for the measurement of bone resorption of proximal regions (cemented enamel junction to alveolar bone crest). Data were submitted to statistical analysis using the Kruskal Wallis and Mann-Whitney tests, with a significance level of 0.05. Rats receiving RESV, β- CD / RESV and placebo received higher bone loss in the presence of ligation (p < 0.05). In the presence of ALN, ligation did not increase bone loss (p > 0.05). In conclusion, we found that the ALN, when compared to itself, in a more acute bone loss, does not seem to accentuate the bone loss

O objetivo desse estudo foi analisar radiograficamente a influência do alendronato e do resveratrol na periodontite induzida pela Porphyromonas gingivalis (P. gingivalis) com e sem ligadura. Cento e vinte ratos foram submetidos à periodontite experimental através de duas técnicas de indução da doença: com ligadura associada à P. gingivalis (LPg) e sem ligadura associada à P. gingivalis (SLPg). Após a doença instaurada, os ratos foram divididos em 4 grupos de acordo com o tratamento: o grupo alendronato (ALN) que recebeu a dose de 2 mg/kg/dia de alendronato; o grupo resveratrol (RESV) que recebeu a dose 10 mg/kg/dia de resveratrol dissolvido em 10 mg/ml de amido solúvel; grupo β- ciclodextrina (β- CD) /RESV que recebeu a dose 10 mg/kg/dia do complexo e grupo controle que recebeu a dose 10 mg/kg/dia de β- CD. As medicações foram administradas via oral por gavagem durante 30 dias. Os animais foram eutanasiados e as peças cirúrgicas da região das maxilas foram radiografadas e submetidas ao software ImageJ ® para a medição da reabsorção óssea das regiões proximais (junção esmalte cemento à crista óssea alveolar). Os dados foram submetidos à análise estatística através dos testes de Kruskal Wallis e Mann-Whitney com o nível de significância estabelecido de 0,05. Os ratos que receberam a administração dos medicamentos RESV, β- CD/RESV e placebo, apresentaram maior perda óssea, na presença de ligadura (p < 0,05). Já na presença do ALN, a ligadura não aumentou a perda óssea (p > 0,05).
