Efeito da suplementação com vitamina D no aumento ósseo vertical em calvária de ratos: análises microtomográficas e histométricas

Resumo: Vitamin D has been studied and tested in bone regeneration with positive but no study evaluated vertical bone augmentation. The objective of this study was to evaluate the effect of vitamin D supplementation on vertical bone augmentation in rat calvarias through computerized microtomography and histometric analysis. Twenty-eight male rats were divided into two groups: Vitamin D Group and Control Group. The animals of the Vitamin D Group received a subcutaneous application of vitamin D at a dose of 87.5 IU, three times a week on alternate days and the animals of the Control Group received subcutaneous application of saline, both starting one week before the surgical procedure until the period of euthanasia. In the calvaria of each animal, two defects of 5 mm of diameter (defect A and defect B). The defect B was filled with particulate autogenous bone and covered with the acrylic resin cap that served as a guide for vertical bone growth. These animals were subdivided into two subgroups for euthanasia at 15 and 60 postoperative days. After that, it was performed computerized microtomography and histometry analyzes. The data were submitted to statistical analysis with 0.05 significance test. The results showed in the microtomographic analysis that the group Vitamin D 15 days presented greater volume of total bone (p = 0.021) and mature bone (p ˂ 0.001) with significant difference compared to the Control group 15 days. In the microtomographic and historical analyzes at 60 days, the Control group presented a significantly higher amount of neoformed bone compared to the Vitamin D group. We can conclude that vitamin D supplementation had a positive effect in favoring a better guided bone healing in its initial stage.

O objetivo deste estudo foi avaliar o efeito da suplementação com vitamina D no aumento ósseo vertical em calvárias de ratos através das análises de microtomografia computadorizada e histometria. Vinte e oito ratos machos foram divididos em 2 grupos: Grupo Vitamina D e Grupo Controle. Os animais do Grupo da Vitamina D receberam aplicação subcutânea de vitamina D, na dose de 87,5 UI, três vezes na semana em dias alternados e os animais do Grupo Controle soro fisiológico, ambos com início uma semana antes do procedimento cirúrgico até o período da eutanásia. Na calvária de cada animal foi realizado dois defeitos de 5 mm de diâmetro (defeito A e defeito B. O defeito B foi preenchido com osso autógeno particulado e coberto com uma tampa de resina acrílica que serviu de guia de crescimento ósseo vertical. Esses animais foram subdivididos em dois subgrupos para a eutanásia com 15 e 60 dias de pós-operatório. Foram realizadas as análises de microtomografia computadorizada e histometria. Os dados foram submetidos a análise estatística (Teste t de Student) com nível de significância 0,05. Os resultados mostraram na análise microtomográfica que o grupo Vitamina D 15 dias apresentou maior volume de osso total (p = 0,021) e osso maduro (p ˂0,001) em relação o grupo Controle 15 dias. Nas análises microtomográfica e histométrica aos 60 dias, o grupo Controle apresentou significativamente maior quantidade de osso neoformado em relação ao grupo Vitamina D.
