Avaliação de células-tronco da bola de bichat associadas à biomaterial bovino e fibrina rica em plaquetas

Resumo: The research of the stem cells has shown very important advances in the area of tissue regeneration. Several research has confirmed that stem cells can be effective in regenerating muscle tissue, heart valves, myocardium and bone tissue. The feasibility of Mesenchymal Stem Cells (CTMs) for subsequent clinical regenerative use has been the focus of many studies. The objective of the present study was to observe the influence of bovine biomaterial associated or not to Rich Fibrin in Platelets (PRF) in the cultivation of MSCs from the Bichat Bola. Three Bichat Ball pairs were used to isolate and culture the CTMs. After the second cell pass, Flow Cytometry and In Vitro Test were performed with the biomaterial and PRF. The amount of 8 x 103 cells were plated in 24-well plates, triplicates of 4 and divided into 4 groups, Control Groups (CTMs only), Bio-Oss / CTMs (5 mg Bio-Oss was inserted into the well within a transwell with 3 μm porosity), Bio-Oss® + PRF / CTMs Group (5 mg of Bio-Oss mixed with 5 mg of fractionated PRF was inserted into the well within a transwell with 3 μm porosity) and Group PRF / CTMs (5 mg fractional PRF was inserted into the well within a transwell with 3 μm porosity). Flow cytometry showed a positive result for CTMs by CD73, CD29, CD10 and CD90, but with alterations in CD34 and vWF markers that have endothelial cell characteristics. Cell viability evidenced that there was cell death by apoptosis, with an average variation of 2.63% to 3.83%. In vitro analyzes showed that MSCs also remained alive and proliferating in all groups during the 25 days. It can be concluded that Bichat's Ball has undifferentiated CTMs and that its association with the bone substitute and / or PRF was feasible.

Várias pesquisas tem confirmado que as células-tronco podem ser eficazes para realizar regeneração de tecido muscular, de válvulas cardíacas, de miocárdio e tecido ósseo. A viabilização de Células-Tronco Mesenquimais (CTMs) para um posterior uso clínico regenerativo tem sido foco de muitos estudos. O objetivo do presente estudo foi observar a influência de biomaterial bovino associado ou não a Fibrina Rica em Plaquetas (PRF) no cultivo de CTMs, provenientes da Bola de Bichat. Foram utilizados três pares da Bola de Bichat para isolamento e cultivo das CTMs. Após a segunda passagem de células, foi realizada a Citometria de Fluxo e a o Teste In Vitro com o biomaterial e com o PRF. A quantidade de 8 x 103 células foram plaqueadas em placas de 24 poços, em triplicatas de 4 e divididas em 4 grupos, Grupo Controle (CTMs apenas), Bio-Oss/CTMs (5 mg de Bio-Oss foi inserido no poço dentro de um “transwell” com 3 µm de porosidade), Grupo Bio-Oss® + PRF/CTMs (5 mg de Bio-Oss misturado com 5 mg de PRF fracionada foi inserido no poço dentro de um “transwell” com 3 µm de porosidade) e Grupo PRF/CTMs (5 mg de PRF fracionada foi inserido no poço dentro de um “transwell” com 3 µm de porosidade). A citometria de fluxo mostrou resultado positivo para CTMs pela porcentagem positiva para os CD73, CD29, CD10 e CD90, porém com alteração dos marcadores CD34 e vWF que tem características de células endoteliais. A viabilidade celular evidenciou que houve morte celular por apoptose, tendo uma variação média de 2,63% a 3,83%%. As análises in vitro mostraram que as CTMs também se mantiveram vivas e em proliferação em todos os grupos durante os 25 dias.
