Resumo: | Medicinal plants have been used for many years to treat and reduce the symptoms of diseases, so it is important that new research with plants be carried out, aiming to evaluate their medicinal effects, and enabling the production of a phytotherapics. The evaluation of medicinal plants, their compounds and medicinal effects that are popularly known is essential, however, the evaluation of their possible immunomodulatory effects such as stimulation of the production of proinflammatory cytokines and increased activity of certain cells of our immune system is extremely relevant. Ocotea puberula, popularly known as cinnamon-guaicá, cinnamon-parda, cinnamonsebo or as guaicá, is part of the Lauraceae family and is used for dysentery, diarrhea and can be indicated for use in cases of eczema, dermatoses, anti-inflammatory effects, inflammatory, analgesic, antioxidant, and stimulant. This work aims to evaluate the immunomodulatory potential of the crude extract of Ocotea puberula. Hot hydroalcoholic extraction was performed to preserve the alkaloids in the extract. The experimental part consists of a total of 160 male Swiss mice weighing approximately 20g that will be divided into 2 experimental groups, Phase 1 and Phase 2, containing 40 and 120 mice in each group, respectively. Phase 1 will consist of evaluating the production and determination of nitric oxide. Phase 2 consists of the evaluation of hematological and immunological biomarkers in vivo. The data obtained from the analyzes were evaluated through analysis of variance ANOVA followed by the Bonferroni test. The oxide determination, the concentration of 50 ng/mL stood out and was used as the basis for phase 2 of this study. In the analysis of phase 2 hematological biomarkers, it was possible to observe changes in all exposed groups. The main change was a significant increase in neutrophils when compared to the control group. The concentration of nitric oxide in the in vivo experiment showed superior results in all exposed groups and the concentration increased with the time of exposure to the extract. These results indicate that the crude extract of Ocotea puberula has immunomodulatory potential. |

These results indicate that the crude extract of Ocotea puberula has immunomodulatory potential.As plantas medicinais vêm sendo utilizadas há muitos anos para tratar e diminuir os sintomas de doenças, portanto é importante que novas pesquisas com plantas sejam realizadas, visando avaliar seus efeitos medicinais e possibilitando a produção de um fitoterápico. A avaliação de plantas medicinais, seus compostos e efeitos medicinais que são popularmente conhecidos é essencial, porém, a avaliação de seus possíveis efeitos imunomoduladores como estimulação da produção de citocinas pró-inflamatórias e aumento da atividade de certas células do nosso sistema imune tem extrema relevância. A Ocotea puberula, conhecida popularmente como canela-guaicá, canela-parda, canela-sebo ou como guaicá, faz parte da família Lauraceae e é utilizada para disenterias, diarreias e pode ser indicada para uso em casos de eczema, dermatoses, efeitos anti-inflamatórios, analgésicos, antioxidantes e estimulantes. Este trabalho tem como objetivo avaliar o potencial imunomodulador do extrato bruto da Ocotea puberula. Foi realizada a extração hidroalcoólica à quente para preservar os alcaloides do extrato. A parte experimental consiste em um total de 160 camundongos Swiss machos de aproximadamente 20g que serão divididos em 2 grupos de experimentos, sendo eles Fase 1 e Fase 2, contendo 40 e 120 camundongos em cada grupo, respectivamente. A Fase 1 consistirá na avaliação da produção e determinação do óxido nítrico. A fase 2 consiste na avaliação de biomarcadores hematológicos e imunológicos in vivo. Os dados obtidos das análises foram avaliados através da análise de variância ANOVA seguido pelo teste de Bonferroni. O nível de significância adotado será de 5%. No experimento de Fase 1 para a determinação de óxido nítrico in vitro, a concentração de 50 ng/mL se destacou e foi utilizada como base para a fase 2 deste estudo. Na análise de biomarcadores hematológicos da fase 2, foi possível observar alterações em todos os grupos expostos. A principal alteração foi um aumento significativo de neutrófilos quando comparado com o grupo controle. A concentração de óxido nítrico no experimento in vivo apresentou resultado superior em todos os grupos expostos e a concentração apresentou aumento conforme o tempo de exposição ao extrato.
