Resumo: Several experiments were carried out to study the behavior of Jales grapevine rootstock in vitro. Explants were obtained from nodal segments cultured in MS medium with 10 µM of BAP. The effect of different culture media, positions of nodal segments, positions and sizes of explant leaf during subculture, and ways of acclimatization was tested. It was concluded that multiplication of Jales grapevine rootstock can be done in free growth regulators half strength MS medium through subculture of established plants in nodal segments with one leaf. Rooting occurs during multiplication, which represents an average index of 3.5 new plants per month. The acclimatization can be made initially in closed containers for three weeks, keeping plants for more four weeks under intermittent mist before taken to open air.

Os explantes foram obtidos a partir de segmentos nodais cultivados em meio de cultura MS com 10 µM de BAP. Foram avaliados meio de cultura, posição do segmento nodal, posição e tamanho da folha do explante durante o subcultivo e aclimatização. Concluiu-se que a multiplicação do porta-enxerto 'Jales' pode ser realizada em meio de cultura MS com a metade da concentração de sais e isento de reguladores de crescimento, por meio do seccionamento das plantas já estabelecidas in vitro em segmentos com uma folha. O enraizamento ocorre durante a multiplicação, que apresenta uma taxa média de 3,5 novas plantas por mês, e a aclimatização pode ser feita em recipientes fechados por três semanas, mantendo-se as plantas por mais quatro semanas em câmara de nebulização, antes de serem levadas ao ar livre.
