Influence of seasonality on flavonoid content, antioxidant potential and toxicity of Doliocarpus dentatus leaf infusion

Resumo: The Doliocarpus dentatus (Aubl.) Standl. is a plant used in treatment of several health problems, however, there are no studies that assess the influence of seasonality on toxicity and on the chemical composition of the infusion this species leaves. Thus, the present study aimed to evaluate the influence of seasonality on toxicity, chemical composition and antioxidant activity of D. dentatus leaf infusion. Yield (EAJAN19) in summer (EAJAN19), autumn (EAMAI18), winter (EAJUL18) and spring (EASET18), were 35.80%; 37.27%; 33.55%; 32.80% respectively. The levels of flavonoids were higher in EAMAI18 (7.20 ± 0.21 mg g-1) and EAJAN19 (6.76 ± 0.06 mg g-1). In the analysis of antioxidant activity, the IC50 values ​​were EAJUL18 (5.50 ± 0.07 µg mL-1), followed by EASET18 (3.72 ± 0.28 µg mL-1), EAJAN19 (2.78 ± 0.31 µg mL-1) and EAMAI18 (1.24 ± 0.54 µg mL-1). For the different seasons, a direct relationship between flavonoid contents and antioxidant activity was observed. In the toxicity test with Artemia salina, as founded in summer, autumn, winter and spring, independent LD50 of 3.48 ± 0.11 mg mL-1; 3.34 ± 0.13 mg mL-1; 3.67 ± 0.11 mg mL-1 and 3.58 ± 0.13 mg mL-1, respectively, showing no toxicity.

Standl., popularmente conhecida como “cipó mata sede”, “cipó de fogo” e “cipó vermelho”, é utilizada para tratar diversos problemas de saúde, entretanto, não há estudos que avaliem a influência sazonal na toxicidade e na composição química da infusão das folhas desta espécie. Assim, o presente estudo visou avaliar a influência sazonal na toxicidade, composição química e atividade antioxidante da infusão das folhas D. dentatus. O rendimento das amostras obtidas no verão, outono, inverno e primavera foram: 35,80%; 37,27%; 33,55%; 32,80%, respectivamente. Os teores de flavonoides foram maiores nas amostras obtidas no outono (7,20±0,21 mg g-1) e verão (6,76±0,06 mg g-1). No potencial antioxidante os maiores valores foram nas amostras obtidas no inverno (5,50±0,07 µg mL-1), seguido da primavera (3,72±0,28 µg mL-1), verão (2,78±0,31 µg mL-1) e outono (1,24±0,54 µg mL-1). No teste com A. salina as amostras obtidas no verão, outono, inverno e primavera apresentaram DL50 de 3,48±0,11 mg mL-1; 3,34±0,13 mg mL-1; 3,67±0,11 mg mL-1 e 3,58±0,13 mg mL-1, respectivamente, não apresentando toxicidade. Assim, amostras sazonais de D. dentatus apresentaram diferenças significativas no rendimento, teor de flavonoides e potencial antioxidante.
