USE OF ORANGE PEELS FOR EXTRACTION OF ESSENTIAL OIL AND EVALUATION OF ITS BIOLOGICAL ACTIVITIES

Resumo: | The expansion of orange production and the installation of agro-industries mitigate the need for waste utilization. This study investigated the use of residual orange peels for essential oil extraction and evaluation of their potential biological activity. The essential oil (OECL) was obtained from the Valência cultivar (Citrus sinensis L.), acquired in the municipality of Jerônimo Monteiro, Brazil. The fresh peels were quantified for moisture and essential oil content. The OECL was characterized regarding its physical (density and refractive index), chemical (qualitative/quantitative composition) and biological (antioxidant activity, phytotoxicity and cytotoxicity) attributes. The peels had 61.5% moisture and 0.9% essential oil, which had a density = 0.8572 g cm-3 and refractive index = 1.4712, having in its composition: Limonene (96.37%), Linalool (2.26%) and β-Pinene (1.37%). Among the biological activities evaluated, the strong antioxidant character stands out, with an effective concentration of 89 mg mL-1 for scavenging 50% of the free radical 2,2-diphenyl-1-picrylhydrazyl. In the in vitro phytotoxicity test, with Lactuca sativa,it was found that 30 mg mL-1 of OECL did not significantly alter the germination process or root growth, but reduced leaflet emission by 90%. In the in vitro cytotoxicity assay, 30 mg mL-1 of OECL did not affect the viability of Saccharomyces cerevisiaecell growth, but reduced its enzymatic activity by 30%. The exploration of OECL appears promising in terms of yield and biological activities, requiring more in-depth studies of selectivity and mechanisms of action. |

Este estudo investigou o uso de cascas residuais de laranja para extração de óleo essencial e avaliação de sua potencial atividade biológica. O óleo essencial (OECL) foi obtido da cultivar Valência (Citrus sinensis L.), adquirida no município de Jerônimo Monteiro, Brasil. As cascas frescas foram quantificadas quanto ao teor de umidade e de óleo essencial. O OECL foi caracterizado quanto aos seus atributos físicos (densidade e índice refração), químicos (composição qualitativa/quantitativa) e biológicos (atividade antioxidante, fitotoxicidade e citotoxicidade). As cascas apresentaram 61,5% de umidade e 0,9 % de óleo essencial, este apresentou densidade = 0,8572 g cm-3 e índice de refração = 1,4712 tendo em sua composição: Limoneno (96,37%), Linalol (2,26%) e β-Pineno (1,37%). Dentre as atividades biológicas avaliadas destaca-se o forte carácter antioxidante, concentração efetiva de 89 mg mL-1 para sequestro de 50% do radical livre 2,2-difenil-1-picrilhidrazil. No teste de fitotoxicidade in vitro, com Lactuca sativa, verificou-se que 30 mg mL-1 do OECL não alterou significativamente o processo germinativo, nem o crescimento radicular, mas reduziu em 90% a emissão de folíolos. Já no ensaio de citoxicidade in vitro, 30 mg mL-1 do OECL não afetou a viabilidade do crescimento celular da Saccharomyces cerevisiae, mas reduziu sua atividade enzimática em 30%.
