Atividade da NADH-redutase de metemoglobina em hemolisado e membranas eritrocitárias de pacientes hansenianos sob tratamento sulfônico

Resumo: We measured in the blood samples of 72, adults leprosy patients, who were ingesting 100 mg of dapsone/day, the levels of erythrocytes, hemoglobin, methemoglobin, sulphone and reticulocytes. NADH-methemoglobin-reductase was measured both in the hemolysate and ghost cells. As a control, identical tests, except for sulphone measure, were applied to blood samples from 72 healthy individuals who did not ingest any oxidant drug. Significant statistical differences regarding the values in erythrocytes, methemoglobin, reticulocytes and activity of NADH- reductase in the erythrocytes ghosts were found among these two groups. The mean enzyme activity in the hemolysate of the leprosy patients and thehealthy individuals has not differed significantly when expressed in UI/g.Hemoglobin/l. The enzyme activity in erythrocytes ghosts from leprosy patients was significantly smaller then that in erythrocytes ghosts from healthy individuals. The mean level of methemoglobin in the leprosy patients was higher than in the control group, probably due to oxidizer action of sulphone. Tends in view we have found smaller levels of enzymatic activity in the erythrocytes membranes of the leprosy patients and coincident levels in the hemolysate compared to controls, we can affirm that probably this is due to an enzymatic displacement from the cellular membrane to the cytoplasm, with the objective of maintaining in the erythrocyte a constant hemoglobinic balance, in spite of the sulphone oxidizer action.

Idênticos testes, exceto dosagem de sulfona, foram efetuados no sangue de 72 pessoas normais, não ingerindo medicamentos oxidantes. Foram encontradas diferenças estatísticas significativas nas variáveis da série vermelha, metemoglobina, reticulócitos e na atividade enzimática da NADH-redutase nas membranas eritrocitárias entre os dois grupos. A atividade enzimática nas membranas eritrocitárias foi inferior e estatisticamente significativa com relação ao grupo-controle. A atividade enzimática da NADH-redutase no hemolisado não apresentou diferença estatística significativa quando feita a correção pela taxa de hemoglobina. O nível de metemoglobina foi superior nos portadores de hanseníase em relação ao grupocontrole, provavelmente devido à ação oxidante da sulfona.
