Interactions between Oxidative Stress, Tumor Staging and Therapy used in Patients with Colorectal Cancer

Resumo: Introduction: The malondialdehyde (MDA) is an important biomarker for the detection of oxidative stress in biological samples. Objective: The aim of the study was to analyze the association between serum levels of oxidative stress markers, tumor staging and therapy in colorectal cancer, and to compare them with healthy individuals. Method: The sample consisted of 40 colorectal cancer patients under chemotherapy and/or radiotherapy, and 80 healthy individuals were recruited in a clinic specializing in oncology, in the period from January to June 2013. Oxidative stress was assessed by lipid peroxidation which was based on thiobarbituric acid reactive species (TBARS). The results were calculated according to a standard curve made with malondialdehyde. The tumor staging of colorectal cancer was obtained from patient’ medical records. Statistical analyzes were performed using Fisher Exact Test and Mann-Whitney test, with p value <0,05 defined as statistically significant. Results: Serum MDA concentration (mM/L) was 4,1 ± 2,9 in the group with cancer and 1,1 ± 0,4 in the group without cancer (p= 0,001). In relation to tumor staging, average MDA was 2,3 ± 1,5 among the group with early stage (II) and 5,4 ± 3,0 between the group with advanced stage (III) (p<0,001). There was no association between serum concentrations of MDA and therapy realized (p>0,05). Conclusion: Serum concentrations of malondialdehyde were higher in the cancer group than in the group without cancer. In addition, high levels of malondialdehyde were associated with the degree of tumor staging.

Objetivo: El objetivo del estudio fue analizar la asociación entre los niveles séricos de marcadores de estrés oxidativo, la estadificación y el tratamiento en el cáncer colorrectal y compararlos con los sujetos sanos. Método: La muestra consistió en 40 pacientes con cáncer colorrectal bajo tratamiento con radioterapia pero no necesariamente recibieron quimioterapia, y 80 sujetos sanos fueron reclutados en una clínica especializada en oncología, entre el período de enero a junio de 2013. El estrés oxidativo se evaluó mediante la peroxidación lipídica que se basaba em las especies reactivas al ácido tiobarbitúrico (TBARS). Los resultados se calcularon de acuerdo con curva estándar hecha con malondialdehído. La estadificación del cáncer colorrectal se obtuvo de las historias clínicas. Los análisis estadísticos se realizaron utilizando el Test Exacto de Fisher y Mann-Whitney, con el valor de p<0,05 definido como estadísticamente significativo. Resultados: La concentración sérica de MDA (mM /L) fue de 4,1 ± 2,9 en el grupo com cáncer y 1,1 ± 0,4 en el grupo sin cáncer (p=0,001). En relación con la estadificación del tumor, el promedio fue de 2,3 ± MDA 1,5 en el grupo con estadificación inicial (II) y 5,4 ± 3,0 entre el grupo con estadificación avanzada (III) (p<0,001). No hubo asociación entre las concentraciones séricas de MDA y terapia realizada (p>0,05). Conclusión: Las concentraciones séricas de malondialdehído fueron mayores en el grupo con cáncer que en el grupo sin cáncer. Método: A amostra foi composta por 40 pacientes com câncer colorretal, recebendo radioterapia e realizando ou não quimioterapia, e 80 indivíduos saudáveis, recrutados em uma clínica especializada em Oncologia, no período de janeiro a junho de 2013. O estresse oxidativo foi analisado através da peroxida..o lipídica, que foi baseada nas espécies reativas ao ácido tiobarbitúrico (TBARS). Os resultados foram calculados de acordo com curva-padrão feita com malondialdeído. O estadiamento do câncer colorretal foi obtido dos prontuários dos pacientes. As análises estatísticas foram realizadas por meio dos testes exato de Fisher e Mann-Whitney, sendo o valor de p<0,05 definido como estatisticamente significante. Resultados: A concentração sérica de MDA (μM/L) foi de 4,1±2,9 no grupo com cancer e de 1,1±0,4 no grupo sem câncer (p=0,001). Em relação ao estadiamento do tumor, a média de MDA foi de 2,3±1,5 entre o grupo com estadiamento inicial (II) e de 5,4±3,0 entre o grupo com estadiamento avançado (III) (p<0,001). Não houve associação entre concentrações séricas de MDA e terapia realizada (p>0,05).
