Análise de biomarcadores circulantes modulados pela terapia de fotobiomodulação em indivíduos com trombose venosa profunda

Resumo: Venous thromboembolism (VTE) is a disease with high morbidity and mortality that includes pulmonary embolism (PE) and deep vein thrombosis (DVT). Deep vein thrombosis is a common disease and has little specific characteristics, which often makes its diagnosis difficult. Photobiomodulation therapy (PBM) has important anti-inflammatory effects such as reduction of chemical mediators such as cytokines, increased generation of nitric oxide (NO) and decreased migration of inflammatory cells. Changes in microRNA expression have recently been reported after PBM therapy. The aim of the project was to analyze the expression transcriptional elements such as messenger RNA (mRNA) and circulating microRNAs in subjects with DVT after irradiation with FBM therapy. Twenty subjects with subacute thrombosis in femoropopliteal territory and 10 healthy subjects as control were studied. DVT subjects were randomly separated into 2 groups of 10 individuals. Of these, only one group received PBM therapy. PBM therapy was applied at wavelength 660 nm. The peripheral blood of all subjects was collected before and after ultrasound and irradiation. The biomarker analyzes were performed with Real-Time PCR, in addition to Western blot analysis and measurement of the total NO concentration. We observed that individuals with DVT have lower expression of eNOS mRNA, higher expression of IL-6 and reduced concentration of NO compared to healthy individuals. However, after PBM therapy, subjects with DVT increased eNOS expression, decreased IL-6 expression, increased total NO concentration and modulated expression of circulating biomarkers such as miR-195 and mir-338. This is the first time that the effect of PBM therapy has been observed in subjects with subacute deep venous thrombosis, modulating transcriptional elements that can contribute to the management of the disease.

A trombose venosa profunda é uma doença frequente e tem como característica sintomas pouco específicos, o que muitas vezes dificulta o seu diagnóstico. A terapia de fotobiomodulação (FBM) exerce efeitos anti-inflamatórios importantes como redução de mediadores químicos como as citocinas, aumento da geração de óxido nítrico (NO) e diminuição da migração de células inflamatórias. Recentemente foram relatadas alterações na expressão de microRNAs após a terapia de FBM. O objetivo deste projeto foi analisar a expressão elementos transcricionais como RNA mensageiro (RNAm) e microRNAs circulantes em indivíduos com TVP após a irradiação com terapia de FBM em comprimento de onda de 660 nm. Foram estudados 20 indivíduos com trombose subaguda em território femoropoplíteo e 10 indivíduos saudáveis como controle. Os sujeitos com TVP foram separados randomicamente em 2 grupos de 10 indivíduos. Destes, apenas um grupo recebeu a terapia de FBM. O sangue periférico de todos sujeitos foi coletado antes e após ultrassonografia e irradiação. As análises de biomarcadores foram realizadas com PCR em Tempo Real, além de Western blot e dosagem da concentração de NO total. Observamos que indivíduos com TVP apresentam menor expressão de RNAm de eNOS, maior expressão de IL-6 e redução da concentração de NO em relação a indivíduos saudáveis. Entretanto, após a terapia de FBM, indivíduos com TVP apresentaram aumento da expressão de eNOS, diminuição da expressão de IL-6, aumento da concentração de NO total e modulação da expressão de biomarcadores circulantes como miR-195 e mir-338.
