Resumo: | Candidiasis is an opportunistic infection caused by Candida albicans. Considering the antifungal agents treatments disadvantages, the Photodynamic Therapy (PDT) based on the concomitant use of a photosensitizer and a light agent, in the oxygen presence, showed to be an interesting alternative. Methylene blue (MB) has been widely used in antimicrobial PDT (aPDT), but due to the lack of protocol standardization, it is not always possible to verify greater efficiency than traditional treatments. The MB can have its activity modulated once its mechanism of action is defined by its aggregation state. In this sense, the identification of the relationships between aggregation, mechanism of action and effectiveness against microorganisms, as well as the establishment of means and formulations that may favor the most effective mechanism, are essential. This study evaluated the aPDT efficacy with a mouthwash formulation with MB in biofilms of in vitro C albicans, with the objective of relating the aggregation state, photochemical mechanism and its effectiveness. In the planktonic culture study, Candida albicans was used to define which parameters would be adopted in the same species biofilm treatment incubation time in the dark, irradiation time and MB concentration. The highest irradiation time (30 minutes) and the lowest MB concentrations (0.001 and 0.002%) were the most efficient parameters. In a second stage, the biofilm was treated in six experimental groups: Physiological solution, no MB mouthwash, MB 0.002 and 0.005% in saline, MB 0.002% and 0.005% in mouthwash. The experimental group MB 0.005% in mouthwash presented lower viability after treatment which makes us to infer that MB in monomers shows greater effectiveness in aPDT and the photochemical mechanism via singlet oxygen is more effective. This research indicates that the formulations development for MB aggregation control, may result in more efficient clinical protocols. |

Considerando as desvantagens de tratamentos com agentes antifúngicos, a terapia fotodinâmica (PDT, do inglês Photodynamic Therapy) baseada no uso concomitante de um agente fotossensibilizador e luz, na presença de oxigênio, se mostra uma alternativa interessante. O azul de metileno (AM), tem sido amplamente utilizado na PDT antimicrobiana (aPDT), mas devido à falta de padronização dos protocolos, nem sempre é possível verificar maior eficiência que os tratamentos tradicionais. O AM pode ter sua atividade modulada uma vez ao seu mecanismo de ação ser definido por seu estado de agregação. Neste sentido, a identificação das relações entre agregação, mecanismo de ação e efetividade contra microrganismos, bem como o estabelecimento de meios e formulações que possam favorecer o mecanismo mais efetivo são essenciais. Este estudo avaliou a eficácia da aPDT com uma formulação de enxaguante bucal com AM em biofilme organizado de C albicans in vitro, com o objetivo de relacionar o estado de agregação, mecanismo fotoquímico e sua efetividade. No estudo em cultura planctônica, Candida albicans foi utilizada para definir quais parâmetros seriam adotados no tratamento de biofilme da mesma espécie: tempo de incubação no escuro, tempo de irradiação e concentração do AM. O maior tempo de irradiação (30 minutos) e as menores concentrações de AM (0,001 e 0,002%) foram os parâmetros que mais eficientes. Em seguida, o biofilme organizado foi tratado com os parâmetros definidos e dividido em seis grupos experimentais: Solução fisiológica, Enxaguante bucal sem AM, AM 0,002% e 0,005% em soro fisiológico, AM 0,002% e 0,005% em enxaguante bucal. O grupo experimental AM 0,005% em enxaguante bucal apresentou menor viabilidade celular pós tratamento, o que nos faz inferir que o AM em monômeros apresenta maior efetividade na aPDT e o mecanismo fotoquímico via oxigênio singlete é mais efetivo.
