Apesar de vários estudos demonstrarem a ação antifúngica do Chá Preto (CP), pouco se conhece sobre a sua atividade antidermatofítica. O presente estudo surge com o objetivo de investigar a atividade do CP dos Açores, em espécies selecionadas de dermatófitos, através de metodologias in vitro. Para tal, avaliou-se a suscetibilidade antifúngica por ensaios de microdiluição; estudou-se o impacto no crescimento, na morfologia e na ultraestrutura dos dermatófitos por técnicas de microscopia (nomeadamente microscopia confocal de fluorescência e microscopia eletrónica de transmissão); quantificou-se ainda dois importantes componentes da parede celular (quitina e glucano). Os resultados mostraram que o CP dos Açores possui atividade antifúngica contra Trichophyton mentagrophytes, Trichophyton rubrum e Microscopum canis, apresenta um efeito sinérgico com anfotericina B, interfere marcadamente com a morfogénese e ultraestrutura do fungo, e conduz a uma fragilidade da membrana citoplasmática. M. canis é a espécie mais suscetível. Também se identificou que a quantidade total de quitina e de glucano permenece inalterada. Além disso, observamos menor intensidade de fluorescência na parede celular da hifa e aumento da intensidade de fluorescência na parede dos septos, após marcação com Calcofluor white, um marcador de fluorescência que se liga à quitina. Isto pode indiciar a presença de compostos bioativos que afetam especificamente as sintases de quitina associadas à síntese de quitina da parede celular da hifa ou dos septos. Em suma, o presente estudo demonstrou que o CP dos Açores possui propriedades antifúngicas, tornando-o um alvo atrativo para o desenvolvimento de uma nova estratégia terapêutica para o tratamento de dermatofitoses.Due to limitations of the current therapeutic options, the development of new and more effective antifungal chemotherapeutic agents or approaches for treating dermatophytosis is clearly necessary. Despite several studies have demonstrated the antifungal action of Black Tea, little is known about its antidermatophytic activity. The present study was designed to investigate the activity of Azorean Black Tea (BT) on selected dermatophytes using an in vitro approach. For that, we assessed the antifungal susceptibility by broth microdilution assay; studied the impact in dermatophyte growth, morphology and ultrastructure using different microscopy techniques (confocal fluorescence microscopy and transmission electron microscopy); and quantified two major cell wall components (glucan and chitin). Results showed that Azorean BT possesses antifungal activity against Trichophyton mentagrophytes, Trichophyton rubrum and Microsporum canis, and markedly interferes with fungal morphogenesis and ultrastructure. It is shown that it synergises with amphotericin B and leads to cytoplasmic membrane fragility. We also found that M. canis was the most susceptible species. We found that the total amount of chitin and glucan remains unchanged. However, it was identified lower fluorescence intensity in the hyphal cell wall and higher fluorescence intensity in the hyphal septa, when using Calcofluor white, a fluorescent probe that binds chitin. This might indicate that there are bioactive compounds in Azorean BT that affect specifically chitin synthases associated either with the synthesis of chitin in the hyphal cell wall or in the septa.
