Um desses processos consiste na tradução do RNA mensageiro (mRNA), um mecanismo que permite à célula responder rapidamente a condições de stress através de modificações na sua maquinaria básica ou através da manutenção da tradução de certos mRNAs em detrimento de outros. Existem vários elementos presentes na região 5’ não traduzida [do inglês, 5’ untraslated region (5’UTR)] do mRNA que são fundamentais para a regulação da sua tradução, tais como as grelhas de leitura abertas a montante da grelha de leitura principal [do inglês, upstream open reading frames (uORFs)], que estão amplamente difundidas pelo genoma, sendo que a sua inserção ou deleção pode levar ao desenvolvimento de doenças. A eritropoietina humana (EPO) é uma hormona glicoproteica envolvida na indução da eritropoiese em condições de hipóxia. Além de desempenhar um papel fundamental na eritropoiese, a EPO também desempenha outros papéis não hematopoiéticos como consequência da sua expressão noutros tecidos para além do rim (onde ocorre ~90% da sua produção) como nos músculos e também no coração e no cérebro, apresentando propriedades de cardio e neuroproteção. Sabe-se que a regulação da expressão do gene da EPO ocorre ao nível da transcrição e do processamento do mRNA, mas também se sabe que ocorre ao nível da tradução através de uma pequena uORF composta por 14 codões. Tal como deduzido através da comparação dos níveis de mRNA e de proteína em estudos anteriores, esta uORF funciona como um inibidor da tradução da grelha de leitura principal [do inglês, open reading frame (ORF)] da EPO sob condições normais. Isto acontece porque o codão de iniciação (AUG) da uORF é reconhecido pela maquinaria de tradução, impedindo que a mesma reconheça e traduza a ORF principal, tratando-se assim, de um regulador negativo da expressão génica. No entanto, em resposta à hipóxia, esta repressão da tradução da ORF é significativamente aliviada, através de um aumento do leaky scanning do AUG da uORF nestas condições. Em concordância, foi demonstrado que em condições de stress, como a hipóxia, ocorre um aumento significativo da síntese da proteína EPO. Tendo em conta todo este conhecimento, neste projeto, avaliámos se a uORF da EPO reprime a tradução da sua ORF principal em células musculares, cardíacas e neuronais e estudámos o efeito da hipoxia neste mecanismo de regulação da tradução. Pudemos observar que em condições normais, a uORF da EPO reprime de facto a tradução da sua ORF principal nas células musculares e cardíacas e que, com a aplicação do stress da hipóxia, ocorre um alívio significativo da repressão da tradução da uORF principal nestas duas linhas celulares. No caso das células neuronais, verificámos que existe uma tendência para que o mesmo se verifique, no entanto, definimos que deverá ocorrer repetição futura das experiências devido a constrangimentos experimentais derivados da linha celular utilizada, não permitindo a obtenção de valores estatisticamente significativos. Assim, o conhecimento gerado neste projeto pode ser usado para entender melhor a regulação da EPO ao nível da tradução e a sua implicação na resposta das células a condições de stress, mais especificamente, à hipóxia, condição essa que ocorre aquando de eventos de isquemia do coração e do cérebro, ou seja, infartos agudos do miocárdio (IAM) e acidentes vasculares cerebrais isquémicos (AVCs).Several key processes lead to genetic expression regulation, essential to maintain and adapt the cells to their needs. One of these processes consists in the messenger RNA (mRNA) translation, which is a mechanism that allows cells to rapidly overcome stress conditions through modification of its basic translation machinery or by maintaining translation of specific mRNAs over others. There are many features of the 5’ untranslated region (5’UTR) of the mRNA that are fundamental for its translational regulation, such as upstream open reading frames (uORFs), that are widely spread on the genome and their introduction or delection can cause disease. Human Erythropoietin (EPO) is a glycoprotein hormone involved in the induction of erythropoiesis under hypoxia. Beyond its role in erythropoiesis, EPO plays several other non hematopoietic roles as a consequence of its expression in other tissues, such as cardio and neuroprotection. It is known that EPO gene expression regulation occurs at the level of transcription and mRNA stabilization but it can also occur at the translational level by an uORF composed by 14 codons. Just as deduced by comparisons of mRNA and protein levels in previous studies, this uORF works as a cis-acting element that represses translation of the EPO main ORF under normal conditions. However, as demonstrated in previous studies, in response to hypoxia, this repression is significantly released, through an enhanced leaky scanning of the uORF. Additionally, it was also demonstrated that stress conditions such as hypoxia, induce a significant increase of EPO protein synthesis. Given all this knowledge, in this work, we intended to evaluate if human EPO uORF represses downstream main ORF translation in muscle, cardiac and neuronal cells and the effect that hypoxia has on this translation regulation mechanism. We could observe that under normal conditions, EPO uORF in fact represses main ORF translation in muscle and cardiac cells and that, with hypoxia induction, a significant relief on the main ORF translation repression occurs in both cell lines. In the case of the neuronal cells, we verified that there is a tendency for the same occurrence, however, we defined that the experiments should be repeated due to experimental constraints caused by the cells used in this work which did not allow us to obtain statistically significant results.
